Regulation of NS-398 on Expressions of VEGF and MMP-9 in Hepatocullar Carcinoma SMMC-7721 Cell Lines

摘要 目的探讨NS-398对肝细胞癌血管生成和侵袭的调节作用。方法NS-398取0、25、50、75、100μmol/L5个浓度组,每个浓度组选取24h、48h、72h三个作用时间点进行检测。采用RT-PCR和流式细胞技术,观察NS-398对人肝癌SMMC-7721细胞株VEGF、MMP-9表达的影响。结果NS-398对SMMC-7721细胞有明显的生长抑制作用。与对照组相比,NS-398能显著抑制SMMC-7721细胞COX-2、VEGF、MMP-9基因和蛋白表达,并呈显著的剂量和时间依赖性关系(P<0.001)。结论NS-398能显著抑制SMMC-7721肝细胞癌中VEGF、MMP-9mRNA和蛋白的表达。 Objective To explore the role of NS-398 in angiogenesis and invasion in hepatocellular carcinoma. Methods For dose-dependent experiments, media containing various concentrations of NS-398 (0, 25, 50, 75 and 100μmol/L) were used. For time course experiments, media containing various concentrations of NS-398 were used, and the cells were harvested daily for 3 days. The expression of VEGF and MMP-9in SMMC-7721 cells were determined by RT-PCR and flow cytometry method by the treatment with NS-398. Results NS-398 significantly inhibited cell proliferation in both time-dependent and dosedependent manner. Compared with control group, N-398 can inhibit COX-2, VEGF and MMP-9 expression in SMMC-7721 cells in a dose and time-dependent. Conclusion This study suggested that NS-398 inhibited the expression of VEGF and MMP-9 mRNA and protein in SMMC-7721 cells.
