血小板保存前添加饱和氢气的作用研究 被引量:5

The effects of adding saturated hydrogen gas to the platelets before platelet preservation

摘要 目的探索氢气在22℃保存血小板中的作用。方法将12份浓缩血小板(40 mL/份)和10份单采血小板(40 mL/份)每份平分为2等份(20 mL/份),随机取1半作为浓缩血小板实验组(n=12)和单采血小板实验组(n=10):加入氢气至饱和;另1半相应作为2个对照组。室温保存5 d后,检测2组血小板计数(Plt)、血小板平均体积(MPV)、血小板体积分布宽度(PDW)、pH值、葡萄糖、乳酸和CD62p表达水平。结果单采或者浓缩血小板22℃保存5 d后,输注氢气组与对照组比较pH:单采血小板7.17±0.26 vs 7.03±0.35(P<0.05),浓缩血小板7.07±0.15vs 6.85±0.34(P<0.05);CD62p:单采血小板17.39±2.88 vs 21.49±4.2(P<0.01),浓缩血小板17.84±2.54 vs21.73±4.14(P<0.01)。结论氢气作为1种选择性抗氧化剂,可减少22℃保存血小板活化,降低血小板保存损伤,提高保存血小板的质量。 Objective To evaluate the effect of hydrogen gas ( H2 ), a selective antioxidant, on platelet preservation. Methods Random donor concentrates (n = 12) and single donor platelets (n = 10) were collected and preserved by the standard methods. Aliquot each sample(40 mL each) for two platelet storage bags(20 mL each). H2 was added to any one of aliquots before preservation. After 5 days of preservation at 22℃, check the value of platelet account (Plt), mean platelet volume ( MPV), platelet distribute width ( PDW), pH, glucose, lactate and CD62p in each platelet sample. Results After 5 days of preservation at 22℃ ,the pH value in both random donor concentrates(7.07 ± 0. 15 vs 6.85 ± 0.34) (P 〈 0.05) and single donor platelets (7.17 ±0. 26 vs 7.03 ±0. 35) (P 〈0. 05) was significantly retained by the addition of hydrogen gas. The expression level of CD62p on platelets preserved with H2 was significantly lower than those preserved without H2 ( single donor concentrates : 17.39 ± 2. 88 vs 21.49 ± 4. 2 ( P 〈 0. 01 ) ; random donor concentrates ( 17. 84 ± 2.54 vs 21.73 ± 4. 14 ) ( P 〈 0. 01 ). Conclusion Hydrogen gas may play an effective role in preserving platelets by inhibiting the activation of platelets and maintain the pH value in the plasma.
