摘要
目的:研究白藜芦醇对胰岛素抵抗大鼠肝脏的影响。方法:将30只SD雄性大鼠随机分为标准饮食组(NC组)、高脂饮食组(HF组)和白藜芦醇治疗组(HR组),每组各10只。对照组采用标准饲料饮食,另两组予高脂饮食,白藜芦醇组从第8周开始给予白藜芦醇灌胃。16周后行高胰岛素-正常葡萄糖钳夹试验,结束后将大鼠处死,观察大鼠胰岛素抵抗情况以及白介素-6(IL-6)和肿瘤坏死因子-a(TNF-α)的表达情况。结果:NC组和HF组大鼠饲养16周后,HF组大鼠的血糖和胰岛素水平较NC组大鼠明显升高,而葡萄糖输注率(GIR)明显降低(P<0.05);3组大鼠血糖、胰岛素含量以及IL-6和TNF-α的免疫组化结果具有明显差异(P<0.05),且水平表现出HF组>HR组>NC组的特点。结论:白藜芦醇可有效降低胰岛素抵抗大鼠的血糖和胰岛素水平,其机制可能是通过调节其IL-6和TNF-α水平所致。
Objective: To study the effects of resveratrol on the liver of rat with insulin resistance. Methods: A total of 30 SD male rats were randomly divided into standard diet group(the NC group),high fat diet group (the HF group)and resveratrol treatment group(the HR group)with 10 rats in each groupThe control group was feed with the standard diet, the other two groups were feed with the high-fat diet, and the resveratrol group was given with resveratrol from the 8th weeks. All rats had high insulinnormal glucose clamps after 16 weeks, and then were sacrificed. Condition of insulin resistance and the expression of IL-6, TNF-α in rats were observed. Results: Comparing with NC group rats, blood glucose and insulin levels of the HF group rats were increased significantly, while the GIR was decreased signifi- cantly after 16 weeks (P〈0.05), which indicated successful insulin resistance rats model. There were significant differences in blood glucose, insulin levels, IL-6 and TNF-α among three groups (P〈0.05), and the level showed that HF group 〉 the HR group 〉 the NC group. Conclusions: Resveratrol can effectively reduce the blood glucose and insulin levels of rats with insulin resistance, it may adjust the level of IL-6 and TNF-α.
