摘要
目的 :测定 10位白血病人 8个时间点血清中VP16浓度 ,并进行药物动力学研究。方法 :HPLC(内标法 ) ,流动相 :甲醇 乙腈 水 (5 0∶10∶40 ,v/v) ,波长 2 5 4nm。结果 :分离良好 ,回收率 >92 % ,日内、日间精密度 <5 % ,药动学参数为二房室开放模型 ,老年人t1/ 2 β有较明显延长。结论 :本法提取简便、灵敏度高 ,精密度高 ,肾功能不良者应减量 ,适用于基层医院VP16的临床监测。
OBJECTIVE:Determine the concentration of VP 16 in serum at eight time points from ten Leukemia′s,and carry out pharmacokinetic studies.METHODS:HPLC(internal standard)was employed,the mobile phase was methanol acetonitrite water(50∶10∶40, v/v ),the wavelength for detection was 254nm.RESULTS:Good spearation and recoveries over 92% were obtained.Coeffcients of variation within a day and between days were under 5%.Pharmacokenitic study indicated that it was two compartment open model with old persons′ t 1/2β extents distinctively.CONCLUSION:This method is simple with high sensitivity and good precision,and is suitable to the determination of VP16 in serum for basic level hospital.
