摘要
目的 建立5个miniSTR基因座的复合扩增系统,提高STR基因座对陈旧骨检材的个体识别效能.方法 新设5个STR基因座的引物,分别用FAM、TAMRA、TET标记D3S3053、D18S853、D12ATA63、D6S474、D9S1122,通过PCR扩增,利用ABI3100xL遗传分析仪进行片段长度分析,对84份河南汉族无关个体血样,以及25例陈旧骨检材进行检测.结果 5个基因座均得到有效扩增,在河南汉族人群中5个miniSTR基因座个体识别能力DP为0.8768~0.9623,杂合度H为0.7454~0.8934,多态信息含量PIC为0.7145~0.8214.5个miniSTR基因座总DP值为0.999 996 72.用Identifil试剂盒及miniSTR技术对25份陈旧骨检材的位点检出率分别为60.25%和84.80%.结论 上述5个miniSTR基因座在河南汉族人群中等位基因分布较好,个体识别能力高,适用于个人识别和亲权鉴定,同时为陈旧骨痕量DNA样本分型提供了一种新的检测方法.
Objective To establish five loci multiplex amplification miniSTR systems, and to improve forensic detection performance of the obsolete bone samples with the STR loci. Methods The new five STR loci( D3S3053 ,D18S853 ,D12ATA63, D6S474, D9S1122) primers were designed and marked by FAM, TAMRA,TET, amplified by PCR and analysized using the ABI3100xL genetic analyzer for fragment length analysis. 84 Henan Hart Nationality blood samples of unrelated individuals, and 25 cases of obsolete bone samples were collected for testing. Results 5 loci were effectively amplified, the 5 miniSTR recognition rate(DP) of individual loci in the Han Nationality population was 0. 8768 - 0. 9623, the heterozygosity (H) was 0. 7454 - 0. 8934, and the polymorphism information content (PIC) is 0. 7145 - 0. 8214. The total 5 miniSTR loci DP was 0. 999 996 72. The detection rate of remaining genetic locus of the 25 obsolete bones was 84.80% by Identifiler kit, and 60.25% by miniSTR method. Conclusion The five miniSTR loci in Han Nationality population in Henan Province is identification and paternity testing, and provides a better distributed and easily recognized as a individual new detection method for DNA samples typing of obsolete bones.
