间质流膏对博莱霉素致大鼠肺纤维化模型PDGF-BB与α-SMA表达影响随机平行对照研究 被引量:1

摘要 [目的]观察间质流膏对博莱霉素致大鼠肺纤维化PDGF-BB与α-SMA表达影响。[方法]使用随机平行对照方法,将96只雄性Wistar清洁级大鼠编号按随机数字表法分为5组,A对照组(12只)、B模型组(28只)、C间质流膏加地塞米松组(28只)、D间质流膏组(28只)、E地塞米松组(28只)。10%水合氯醛大鼠腹腔注射麻醉,额镜直视下气管插管,气管滴入博来霉素生理盐水0.1mL(5mg/kg),滴注后立即将大鼠直立旋转,使药液在肺内分布均匀,复制肺纤维化模型。A、B组生理盐水腹腔注射0.3ml/d。C组间质流膏灌胃(党参24g,黄芪18g,白术、茯苓各12g,沙参20g,水蛭研末冲6g,蜈蚣研末冲2条,丹参21g,桃仁、川芎、地龙各15g,黄芩12g,白花蛇舌草、薏苡仁各30g,蒲公英24g,浙贝15g,莪术12g,山慈菇21g,甘草12g,连翘、鱼腥草各18g)10mL/kg大鼠体重(生药4.6g/10mL),地塞米松腹腔注射1mg/kg,1次/d。D组间质流膏灌胃(生药4.6g/10mL)10mL/kg大鼠体重,1次/d。E地塞米松腹腔注射1mg/kg,1次/d。连续干预4周(28d)。实验第28天A组脱颈椎处死大鼠3只,其他组分别处死大鼠7只,暴露气管,向大鼠右肺灌注10%甲醛溶液,使胸膜展平后将右肺浸入10%甲醛溶液中固定。常规石蜡包埋,切片(4μm)行HE和免疫组化染色。观测光镜下肺组织病理细胞、肺组织PDGFBB、α-SMA。[结果]B模型组肺泡结构破坏,肺泡壁显著增厚,肺间质纤维化瘢痕形成,毛细血管腔闭塞;C间质流膏加地塞米松组肺组织呈轻中度肺纤维化改变,病变范围局限,肺泡间隔胶原沉积较模型组减少;D间质流膏组与激素组比较肺纤维化程度明显减轻,较正常组略有改变。E地塞米松组肺纤维化改变不明显。肺纤维化程度由轻到重依次为:A组>C组>D组>E组>B组。PDGF-BB值,与A组比较,B组各时间段均明显增高(P<0.05);7d组间比较(P<0.01),D组与E组(P>0.05);14d组间比较(P<0.01),D组与E组(P>0.05);21d组间比较(P<0.01),两两比较均(P<0.05);28d组间比较(P<0.01),两两比较均(P>0.05)。[结论]间质流膏可明显降低博莱霉素致大鼠肺纤维化模型PDGF-BB与α-SMA表达。 [ Objective ] To study the influence on PDGF-BB and α-SMA levels by Jianzhi-liquid on Bleomycin-induced Idiopathic interstitial pneumonia in rats models. [ Methods ] The use of random parallel control method, the number of 96 male Wistar rats of clean grade were randomly divided into 5 groups, A group ( 12 rats ), B model group ( 28 rats ), paste plus dexamethasone group C interstitial flow ( 28 rats ), paste group D interstitial flow ( 28 ), dexamethasone group ( E 28 rats ) . Anesthesia by intraperitoneal injection of rats with 10% chloral hydrate, amount of endoscopic tracheal intubation, intratracheal instillation of bleomycin 0.1mL normal saline (5mg/kg), drip immediately after the rats upright spin, so that the liquid medicine is evenly distributed in the lungs, pulmonary fibrosis model replication. A, group B with normal saline intraperitoneal injection of 0.3ml/d. Group C Jianzhiliugao gavage (24g dangshen, astragalus, Poria Atractylodes 18g, each 12g, Radix 20g, leech at the end of the research at 6g, the centipede powder at 2, 21g salvia, chuanxiong,peach kernel, earthworm each 15g, 12g of Radix Scutellariae, spreading hedyotis herb, coix seed 30g, dandelion 24g, Fritillaria thunbergii 15g 12g, zedoary, mountain arrowhead 21g, licorice 12g, forsythia, tterba Houttuyniae each 18g ) the body weight of 10mL/kg rats ( 4.6g/10mL ), intraperitoneal injection of dexamethasone lmg/kg, 1 /d. Group D Jianzhiliugao gastric perfusion ( 4.6g/lOmL ) the body weight of 10mL/kg rats, 1 /d. E intraperitoneal injection of dexamethasone lmg/kg, 1 /d. Continuous intervention for 4 weeks ( 28d ) . Experiment twenty-eighth days in group A, 3 rats were sacrificed by cervical dislocation, the other group were killed 7 rats exposed to rats, trachea, right lung perfusion of 10% Formaldehyde Solution, the pleural flattened the right lung in 10% Formaldehyde Solution fixed. Routine paraffin embedding, slicing ( 4μm ) for HE and immunohistochemical staining. Observation under light microscope, lung cells, lung tissue PDGF-BB, α-SMA. [ Results ] In B model group, alveolar structural damage, alveolar wall thickening, interstitial fibrosis and scar formation in lung, capillary lumen occlusion; C Jianzhiliugao plus dexamethasone group were mild to moderate pulmonary fibrosis, alveolar septal lesions confined, collagen deposition was lower than that of model group; D Jianzhiliugao group and the hormone group level pulmonary fibrosis was significantly reduced, changed slightly compared with normal group. E dexamethasone group did not change significantly in pulmonary fibrosis. The degree of pulmonary fibrosis from light to heavy as follows : A group, C group, D group, E group, B group. The value of PDGF-BB, compared with A group, B group each time increased significantly ( P〈 0.05 ) ; compared with 7d group ( P〈 0.01 ), D group and E group ( P 〈 0.05) ; compared with 14d group ( P〈 0.01 ), D group and E group ( P〈 0.05 ) ; comparison of 21d group ( P〈 0.01 ), two two were ( P〈 0.05 ) ; compared with 28d group ( P〈 0.01 ), two two were ( P〈 0.05 ) . [ Conclusion ] Jianzhi-liquid could effectively low the expressions of PDGF-BB and ot -SMA, and might be one of the mechanisms of Jianzhi-liquid treatment of pulmonary fibrosis.
