Sox9和Ⅱ型胶原蛋白在人腰椎间盘退变中的表达及意义 被引量:15

摘要 目的探讨Sox9和Ⅱ型胶原蛋白(Col2al)在人类腰椎间盘退变中的表达及意义。方法应用免疫组织化学在退变和正常腰椎间盘组织中分别检测Sox9和Ⅱ型胶原蛋白的表达,并分析其相关性。结果实验组Sox9染色阳性率为20.0%(6/30),对照组Sox9染色阳性率为76.7%(23/30),两组间染色阳性率有显著性差异(P<0.05)。实验组Col2al染色阳性率为33.3%(10/30),对照组Col2al染色阳性率为83.3%(25/30),两组间染色阳性率有显著性差异(P<0.05)。退变腰椎间盘中6例Sox9阳性者中,仅1例Col2al表达阴性,而在10例Col2al表达阳性者中,5例Sox9表达阴性,二者同时阳性5例,同时阴性表达19例。退变椎间盘中Sox9和Col2al的表达呈显著正相关(P<0.05)。结论 Sox9和Ⅱ型胶原蛋白表达降低与腰椎间盘退变有关,Sox9可能通过下调Col2al的表达,共同参与腰椎间盘退变。 Objective To study expressions of Sox9 and Col2a1 in human degenerated lumbar intervertebral disk. Methods Immunohistochemestry was used to evaluate expression of Sox9 and Col2a1 in human degenerated and nomal lumbar intervertebral disk. Results The positive immunostaining rate of Sox9 was 20.0%(6/30) in experimental group and 76.7% (23/30) in control group. There were significant differences(P 〈 0.05) between two groups. The positive immunostaining rate of Col2a1 was 33.3% (10/30) in experimental group and 83.3% (25/30) in control group.There were significant differences(P 〈 0.05) between two groups.In the 30 degenerated lumbar intervertebral disks, 6 cases of Sox9 positive expression included 1 case of Col2a1 negative. 10 cases of Col2a1 positive expression included 5 cases of Sox9 negative expression. 5 cases were both positive while 19 cases were both negative. There was a positive correlation between the expressions of Sox9 and Col2a1(P 〈 0.05). Conclusion Decrease of Sox9 and Col2a1 is related to the degeneration of lumbar intervertebral disk. Sox9 may downregulate Col2a1 directly,resulted in degeneration of lumbar intervertebral disk.
