摘要
目的 探讨膜联蛋白A2在结直肠癌中的磷酸化水平及其与临床病理特征及肿瘤预后的相关性.方法 分析2006年8月至2007年7月在第二军医大学附属长海医院肛肠外科进行治疗的183例散发性结直肠癌患者的临床及随访资料.建立患者肿瘤标本组织芯片,对磷酸化膜联蛋白A2的水平进行免疫组织化学检测并对结果进行统计分析.结果 肿瘤组织中磷酸化膜联蛋白A2的水平高于非恶性的对照组织（P＜0.01）.患者肿瘤组织中较高的磷酸化膜联蛋白A2水平与T分期（P＜0.01）、N分期（P＜0.05）、肝转移（P＜0.01）、TNM分期（P＜0.01）、Duke分期（P＜0.01）、复发（P＜0.01）、肿瘤致死（P＜0.05）密切相关.生存曲线分析显示肿瘤组织中磷酸化膜联蛋白A2水平较低的患者其未复发率（P＜0.01）和总体生存率（P＜0.05）均高于磷酸化膜联蛋白A2水平较高的患者.多因素分析显示肿瘤组织中磷酸化膜联蛋白A2的水平可以作为衡量患者未复发率（P＜0.05）和总体生存率（P＜0.05）的独立因素.结论 结直肠癌患者肿瘤组织中磷酸化膜联蛋白A2的水平与肿瘤分期和临床特征密切相关.
Objective To investigate an insight into the putative role of phosphorylation-annexin A2 （p-ANXA2） in tumor stage and prognosis of colorectal adenocarcinomas.Methods A total of 183 patients with sporadic colorectal adenocarcinomas （CRC） enrolled in this study and the tumor tissue samples were obtained by surgical resection with corresponding nonmalignant controls.Then the level of p-ANXA2 in tumor tissues and controls were investigated by immunohistochemical staining and data was statistically correlated with pathologic parameters and clinical outcome.Results Immunostaining showed that the level of p-ANXA2 in tumors was higher than controls （P ＜ 0.01）.Univariate analysis disclosed increased p-ANXA2 level correlated with T stage （P ＜ 0.01）,N stage （P ＜ 0.05）,liver metastasis （P ＜ 0.01）,TNM stage （P ＜ 0.01）,Duke stage （P ＜ 0.01）,recurrence （P ＜ 0.01）,cancer-related death （P ＜0.05）.Survival curves disclosed statistically significant correlation of high level of p-ANXA2 with increased tumor recurrence-free probability （P ＜ 0.01） and overall survival （P ＜ 0.05）.Multivariate analysis disclosed that p-ANXA2 level significantly correlated with recurrence-free probability （P ＜ 0.05） and overall survival （P ＜ 0.05） independently.Conclusion The level of p-ANXA2 in tumor was related to the tumor stage and clinical outcome of CRC.
