摘要
目的观察钙磷脂结合蛋白Ⅱ(Annexinll)在人肝细胞癌(HCC)组织中的表达。方法采用荧光实时定量聚合酶链反应(Real—timePCR)方法检测33例HCC患者肝癌组织和配对的癌旁肝组织以及11例正常肝组织中AnnexinⅡmRNA表达;利用免疫组织化学方法检测104例HCC组织和配对的癌旁肝组织中AnnexinⅡ蛋白的表达。结果肝癌组织中AnnexinⅡmRNA表达量高于癌旁肝组织和正常肝组织,是癌旁肝组织的3.38倍(P〈0.01),是正常肝组织的4.42倍(P〈0.01);AnnexinII蛋白表达明显高于配对的癌旁肝组织(P〈0.05)。年龄≤40岁的患者AnnexinⅡmRNA表达水平是年龄〉40岁患者的1.875倍(P〈0.05)。结论AnnexinII在人HCC患者肝癌组织中表达上调,可能成为肝细胞癌临床诊断的新的候选基因。
Objective To investigate the Annexin II expression levels in hepatocellular carcinoma (HCC). Methods SYBR green I Real-time polymerase chain reaction ( Real-time PCR) was used to detect Annexin II mRNA expression in 33 paired HCC tissues and peritumoral liver tissues and in 11 normal liver tissues. Annexin II protein was detected by immunohistochemcal staining in tissue chips of 104 cases of HCC tissues and adjacent noncancerous liver tissues. Results Annexin 1I mRNA expression in HCC tissues was 3.38 times higher than that in their paired noncancerous liver tissues (P 〈 0. 01 ) and was 4. 42 times higher than that in normal liver tissues ( P 〈 0. 01 ). Annexin II mRNA expression in peritumoral liv- er tissues was 1.94 times higher than that in normal liver tissues ( P 〈 0.05 ). The protein expression of Annexin IX in HCC tissue was significantly higher than that in adjacent noncancerous liver tissues (P 〈 0. 05 ). Annexin II mRNA level in HCC tissues of age ≤40 years was 1. 875 times higher than that in age 〉 41 years ( P 〈 0.05 ). Conclusion Upregulation expressions of Annexin II may serve as a potential diagnosis biomarkers for HCC.
