摘要
目的 探讨三氧化二砷 (As2 O3)诱导肝癌HepG2细胞凋亡作用。 方法 应用普通光学显微镜、荧光显微镜和流式细胞仪观察As2 O3 对HepG2细胞株的形态学改变和诱发凋亡率。结果 As2 O3 作用于细胞后 ,可看到较典型的细胞凋亡形态学改变 ,细胞体积缩小 ,染色质固缩成斑块状 ,呈半月型紧贴于核膜周边 ,核碎裂 ,凋亡小体形成等。AO/EB荧光染色法显示 ,细胞凋亡率为 4.3 %~ 9.1%。As2 O3 诱导HepG2细胞凋亡作用呈时间依赖性 ,最佳浓度为 2 μmol/L。流式细胞仪DNA直方图上呈现典型的亚二倍体凋亡峰。As2 O3 主要作用于细胞周期的G2 /M期。结论 As2 O3 具有诱导HepG2细胞凋亡作用 ,存在治疗肝癌的潜在价值。
Obiective To explore the effect of arsenic trioxide (As 2O 3) on induction of apoptosis of liver cancer cell lines HepG2. Methods The morphologic changes and apoptosis rate of HepG2 cell induced by As 2O 3 were observed with fluorescent microscopy and flow cytometry.Results After 24 hours of exposure to the drug, HepG2 exhibited typical morphologic changes of apoptosis ,including shrinkage of cell,condensation of chromatin, breakage of neuclear and formation of apoptotic bodies.The apoptotic rate of HepG2 was 4.3%~9.1% by fluorescent microscopy assay. The best concentration of As2O3 was 2 μmol/L. A typical subdiploid peak before G1 phase was observed by flow cytometry. As2O3 mainly acts in G2/M phase. Conclusion As 2O 3 can induce apoptosis of HepG2 and it has a potential value of clinical therapy on liver cancer.
