摘要
目的 探讨氧化砷(As2O3)对结肠癌细胞SW480的增殖抑制作用。方法 应用台盼蓝拒染法、MTT比色法及细胞克隆形成试验,研究As2O3对结肠癌细胞的增殖抑制和细胞毒作用。结果 As2O3能显著抑制结肠癌细胞的生长,5μmol/LAs2O3抑制强度明显强于1.25mol/L、1.25μmol/L和5μmol/L As2O3作用后克隆形成率分别为(9.6±2.1)％和(1.7±0.9)％,明显低于对照组的(48.2±6.7)％(P<0.01)。As2O3对结肠癌细胞有较强的细胞毒作用,且呈剂量和时间依赖性,其对结肠癌细胞的杀伤率显著高于5-氟尿嘧啶(5-FU)。As2O3与5-FU存在协同效应,联合应用后,对结肠癌细胞的杀伤率明显升高。结论 As2O3具有明显抑制结肠癌细胞增殖的作用,存在治疗结肠癌的潜在价值。
Objective To study the effect of arsenic trioxide (As2O3) on inhibition of proliferation of human colon cancer lines SW480. Methods The morphologic changes,proliferation ability, colony-forming rate of SW480 treated with As2O3 were studied by light microscopy, growth curve, trypan blue,and colony-forming assay.The cytotoxicity of As2O3 on SW480 was determined by MTT assay.Results As2O3 inhibited significantly the growth of SW480. Colony-forming rate of SW480 treated with 5 μmol/L As2O3[(1.7 ± 0.9)% ] was lower than that with 1.25μmol/L As2O3[(9.6±2.1)%, P <0.01 ],and much lower than that in control group [(48.2±6.7)%, P <0.01]. As2O3 had a remarkable cytotoxic effect on SW480 dose-and time-dependendy. The cytotoxicity of As2Q3 on SW480 was remarkably higher than that of 5-FU. As2O3 combined with 5-FU enhanced the cytotoxic effect. Condusion Arsenic trioxide significantly inhibits proliferation of SW480 and has a potential value clinical therapy of colon cancer.
