摘要
目的 探讨原发性高血压 (EH)时人体血液系统 L -精氨酸 (L - Arginine,L - Arg)载体转运功能的改变 .方法 EH患者 12例及健康成年人 16例 ,提取血小板 (Pt)和红细胞(RBC) ,分别测定 Pt和 RBC的 L- Arg转运的动力学特征 .结果 EH患者 Pt和 RBC对 3H- L - Arg转运呈高亲和性载体转运方式 ,在不同浓度点 EH患者的 3H- L- Arg转运率均比对照组明显低下 (P<0 .0 1) .Eadie- Hofstee Plot分析 ,EH患者Pt L - Arg的最大转运速率 (Vmax)为对照组的 79% (P <0 .0 5 ) ,而 Pt3H- L - Arg转运的米氏常数值 (Km)无明显差异 .RBC的 L - Arg转运总能力 Y+ 载体转运系统的转运能力明显低于正常组 (P<0 .0 1) ,而 Y+ L载体转运系统的转运能力无明显改变 .L- Arg总转运的最大速率与 Y+载体转运系统的最大速率分别较对照组降低 36 .8%和 42 .5 % (P<0 .0 1) ,各组之间 Km 无明显差异 .结论 EH患者 Pt和 RBC L -
AIM To investigate the changes of L arginine (L Arg)transportation kinetic characteristics in erythrocytes (RBC) and platelets (pt) of patients with essential hypertension (EH). METHODS 12 EH patients were studied with 16 normotensive people as control. The RBC and Pt were prepared and the transportation of L Arg was measured by using the method described by Ghigo and Deliconstantins. RESULTS In EH patients, the ability of Pt L Arg transportation was lower than control and V max decreased by 21% ( P <0.01). In RBC, Saturable influx of L Arg was mediated by teh classical cationic amino acid transport system Y + and Y +L system with high affinity. Under zero trans condition, the ability of L Arg transportation in EH patients was lower than control. The V max for total L Arg transport and via system Y + decreased 36.8% and 42.5% respectively compared with control ( P <0.01). But V max for L Arg transport via system Y +L was not affected by EH. There was no obvious difference between the groups. CONCLUSION There might be disturbance of L Arg transportation in human erythrocytes and platelets of patients with EH.
