摘要
目的 :探讨福斯可林 (forskolin)或 /和 3 异丁基 1 甲基黄呤嘌 (IBMX)玻璃体内注射对视网膜节细胞再生的作用。材料和方法 :采用逆行示踪标记术 ,研究外周神经缝接的对照组和外周神经缝接后 ,玻璃体内注射forskolin和 /或IBMX的实验组视网膜节细胞再生情况。结果 :(1 )对照组 (AG)术后 4、6周 ,每个视网膜再生的节细胞数分别为 :1 2 60± 1 3 7个和 95 7± 1 75个 ;(2 )在上述两个时间点上各实验组每个视网膜再生的节细胞数分别为 :①forskolin组 (AG +F) :1 80 3± 64 8个和 1 0 68± 3 88个 ;②IBMX组 (AG +I) :2 2 84± 1 0 0 5个和 1 0 0 2± 1 5 7个 ;③forskolin和IBMX联合应用组 (AG +I+F) :480 4± 3 75个和 3 1 61± 60 5个 ;其中 ,AG +I+F组同AG ,AG +F ,AG +I组相比在这两个时间点上均存在显著性差异 (P <0 .0 0 1 )。结论 :研究结果提示联合应用forskolin和IBMX可大幅提高视网膜节细胞的再生。
To investigate the effects of forskolin and/or IBMX on axotomized retinal ganglion cells (RGCs) in adult hamsters. Methods: Fluorescent retrograde labeling method was used to measure the quantity of RGCs regeneratin in control and experimental animals treated with forskolin and/or IBMX 4 and 6 weeks after surgery. Results:(1) In control(AG) retinas, the mean number of regenerating RGCs were 1260±137/retina and 957±75/retina at 4,6 week after surgery, respectively, (2) The mean number of regenerating RGCs in AG+F group at 4 week and 6 week were 1803±648/retina and 1068±388/retina, respectively.(3) In AG+I group, the mean number of regenerating RGCs at 4 week and 6 week were 2284±1005/retina and 1002±157, respectively.(4) The mean number of regenerating RGCs in AG+I+F were 4804±375/retina and 3161±605/retina at 4 week and 6 week after surgery, respectively. They were significantly different from groups of AG, AG+F and AG+I(P<0.001). Conclusion: Our results indicate that forskolin combined with IBMX has significant effects in promoting the regeneration of RGCs after axotomy.
