摘要
目的探讨高碘环境下体外培养的甲状腺滤泡上皮细胞分泌 IL-1α、 IL-6的变化,以及与自身免疫性甲状腺疾病(AITD)的关系。方法应用原代细胞培养技术,术中取甲状腺腺瘤旁组织进行培养,然后加入不同浓度的碘继续培养48h,用双抗体夹心 ELISA 法测定上清液中IL-1α、IL-6的含量。结果当碘浓度为 0.15 μg/ml,0.75 μg/ml时,IL-1α的分泌量增加(与对照组相比 P<0.01),当碘浓度为 0.7μg/ml、3.75 μg/ml时,IL-6的分泌量增加(与对照组相比 P<0.01)。结论高碘可刺激甲状腺细胞分泌更多的 IL-1α、IL-6。高碘在AITD发病过程中起重要作用。 IL-1α、IL-6可做为碘损伤甲状腺细胞的诊断指标。
Objective To study the effects of high iodine on secretion of interleukin-1α (IL-1α ) and interleukin-6 (IL- 6) by human thyrocytes in primary culture and the relation between iodine and autoimmune thyroid diseases (AITD). Meth- ods Thyrocytes were cultured for 48 hours in different iodine concentration medium. The contents of IL-1α, IL-6 in super- nate were determined by ELISA. Results The results showed that there were high contents of IL-1 α in supernate when the iodine concentrations were 0.15μ g/ml and 0.75 μg/ml, ad well as those of IL-6 when foe iodine concentration were 0.75μg/ml and 3.75μ g/ml. (compared with the empty control group. P<0.01). Conclusions High iodine can stimulate the thyrocytes to secrete excessive IL-1 α and IL-6, and high iodine plays a very important role in process of AITD onset. IL- 1α and IL-6 may he the indexes to judgement of thyroid injury.
