摘要
目的研究了低温冻结和液态冷藏条件下,海藻糖对血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、γ—谷氨酰转肽酶(γ-GT)、碱性磷酸酶(ALP)的活性保护作用,并探讨了海藻糖对酶的最佳保活浓度,以提供一种多酶质控品的良好稳定剂。方法采用酶动力学方法分别在30d、60d、120d、240d、360d分别测定四种酶活力,并计算酶的保活率(%)。结果在360d时,海藻糖保护组的四种血清酶的平均保活率为93.7%,而无海藻糖保护组的四种血清酶的平均保活率为67.7%,海藻糖组的保活率是无海藻糖组的1.38倍,尤其是海藻糖对ALT的保活作用尤为显著,其保活率是对照组的2.55倍。海藻糖对酶的保活最佳浓度为0.1~0.25mol/L之间,低于0.1mol/L或高于0.3mol/L则保活效果不佳。同时对液态下海藻糖的保活效果作了时效观察,同样对ALT有较明显的保护作用,保活率是其对照组的2.06倍。结论通过实验研究证实了海藻糖对酶在冻结状态下具有保活作用,是酶蛋白的良好的稳定剂。
Objective To study the trehalose protection of four serum enzymes at frezzeing and refrigeration and dex-plore the best activity concentration of enzymes under trehalose protection. Toelevelop a good stabilizer of multienzymes QC material. Methods The four enzymes activily were measured with kinetic at 30days, 60days, 120days, 240days, 360days and ealculate enzymes protection activity rate ( %) . Results At 360days. the trehalose group protection of four serum en zymes average PAR 67.7 % . Trehalose group is 1.38 times as large as mon - trahalose group. Especially of ALT activity ex -taaordinary remarkable under trahalose protection.Trehalose group is 2.55 time as large as nontrchalose group.The best activity concentration of enzymes between 0.1 to 0.25 mol/L, Conclusions Laboratory investigate evidence the trehalose protection enzymes under freeze.The trehation is a good stubilizer of enzyemes protien.
