The Activity of SOD and its Isozymes & Thermostability in Reguminosae Seeds

摘要 13种豆类种子超氧物歧化酶 (SOD)活性、同工酶及热稳定性研究的结果表明 :豆类种子含有较高的SOD活性 ,每克鲜重含 5 0 0~ 12 0 0酶单位 ,其中绿豆、赤豆、蚕豆、大豆种子SOD活性都在 10 0 0U/g以上 .豆类种子一般含有Cu·Zn -SOD和Mn -SOD两种类型的同工酶 ,以前者为主 ,其中Mn -SOD活性一般占总活性的10 % ,粉青豆Mn -SOD活性最高 ,为 198U/g .聚丙烯酰胺凝胶电泳 (PAGE)同工酶谱显示多数受试种类含 1条Mn -SOD区带和 3~ 6条Cu·Zn -SOD区带 . Superoxide Dismutase in seeds of 13 kinds from Reguminosae were assayed.Results indicated that all the members in this family had high SOD activities, which ranged from 500 to 1?200?U/g, and in Green Gram,Granisia,Bread bean and Soybean ,they were all above 1?000?U/g. Reguminosae seeds generally contained two kinds of SOD isozymes:Cu·Zn SOD and Mn SOD.The former was the main and the latter contained about 10% of the total activity. Glycine max (L.) Merr contained the most abundant Mn SOD activity which was 198?U/g and was about 29.2% of the total activity. The PAGE band in most of the assayed members contained 1-band of Mn SOD and 3~6 bands of Cu·Zn SOD. SOD in the variety of high Mn SOD activity had a good thermostability.
