摘要
目的研究Livin、PTEN及基质金属蛋白酶-9(matrix metauoproteinase-9,MMP-9)在视网膜母细胞瘤(retinoblastoma,RB)中的表达和临床意义,并探讨三者之间的相关性。方法对56例RB组织标本进行Livin、PTEN及MMP-9蛋白表达的免疫组织化学染色,以正常视网膜组织标本10例作为正常对照,采用免疫组织化学法检测56例RB组织中及10氉例正常视网膜组织中的Livin、PTEN及MMP-9的阳性表达情况,探讨3种因子在正常视网膜及RB中的表达及相关性。结果Livin蛋白在RB中阳性表达,阳性表达率为57.1%,但其在正常视网膜组织中不表达(P<0.05);PTEN蛋白在正常视网膜表达,阳性率达100.0%,而在RB组织中表达率仅为55.4%,差异有统计学意义(P<0.05);RB组织中MMP-9阳性表达率高达50.0%,在正常视网膜组织中不表达,两者比较差异有统计学意义(P<0.05)。PTEN、Livin及MMP-9的蛋白表达与RB分期、病理学分型及伴视神经浸润有关(均为P<0.05),与性别及年龄无关(均为P>0.05)。PTEN和Livin的表达呈显著负相关(r=-0.370,P<0.05),与MMP-9的表达也呈显著负相关(r=-0.310,P<0.05);而Livin和MMP-9在RB中的表达呈显著正相关(r=0.450,P<0.01)。PTEN表达与MMP-9、Livin共表达存在负相关(r=-0.542,P<0.01)。结论 Livin、PTEN及MMP-9可作为判断RB恶性程度和预后的指标,在RB的早期诊断中有重要作用。
Objective To study the expression of Livin,PTEN and matrix metal- loproteinase-9 （ MMP-9 ） and discuss their correlation in retinoblastoma （RB）. Meth- ods The positive expression of Livin, PTEN and MMP-9 in 55 RB tissue samples and 10 normal retinal tissue specimens （normal control） was observed by immunohisto- chemical staining,the correlation of three factors was discussed. Results Livin pro- tein did not express in normal retinal tissue, but positive expressed in RB tissues, the positive expressed rate was 57.1% , there was statistical difference （P 〈 0.05 ）. The posi- tive expression rates of PTEN protein was 100% in normal retinal tissue and significant- ly higher than 55.4% of positive expression in RB tissues, there was statistical differ- ence （P 〈 0.05 ）. MMP-9 protein did not express in normal retinal tissue, but positive ex- pressed in RB tissues,the positive expressed rate was 50% , there was statistical differ- ence （P 〈 0.05 ）. The positive expression of PTEN, Livin and MMP-9 protein was corre- lated with clinical stages, histopathological type and optic nerve infiltration （ all P 〈 0. 05 ）, but not correlated with age and gender （ all P 〉 0.05 ）. Expression of PTEN pro- tein was negatively correlated to both that of Livin and MMP-9 （r = -0.370, -0. 310,all P 〈 0.05 ）. Expression of Livin and MMP-9 in RB tissues was positively correlated （ r = 0. 450,P 〈0.01 ）. Expression of PTEN was negatively correlated with co-expression of MMP-9 and Livin（r = -0. 542,P 〈0.01）. Conclusion Livin,PTEN and MMP-9 are the important indexes in judging the malignant degree and prognosis of RB, which play the key role in early diagnosis of RB.
