摘要
目的探讨基质金属蛋白酶(MMP-2)在糖尿病视网膜病变(DR)发生发展中的作用及其机制.方法用链脲佐菌素(STZ)诱导苍鼠糖尿病模型,提取视网膜中总RNA,半定量RT-PCR观察视网膜中MMP-2mRNA表达情况,HE染色观察光镜下视网膜的形态特征及变化,血清学检测苍鼠血清甘油三酯(TG),总胆固醇(TC)和电化学发光法检测胰岛素水平等变化.结果糖尿病苍鼠不仅表现为高血糖,还表现为高TG血症.视网膜中MMP-2mRNA表达呈升高趋势,与正常对照组相比于造模后16wk末时即差异具有统计学意义(P<0.05).结论此为研究基质金属蛋白酶参与DR的发病机制提供了依据.
AIM: To explore the role of matrix metalloproteinase-2 (MMP-2) in the development of diabetic retinopathy (DR). METHODS: Hamsters were used to establish diabetic models by Streptozotocin (40 mg/kg). Total RNA of retina was collected. The expression of MMP-2 mRNA in retina were observed by using semi-quantitative RT-PCR. Morphologic characteristics and changes of retina was observed by HE staining. The changes of TG, TC were tested serologically and insulin was tested by electrochemiluminescence. RESULTS: Diabetic hamster displayed not only hyperglycaemia, but also hyperlipemia. The expression of MMP-2 mRNA in retina showed a increasing tendency. CONCLUSION: MMP-2 may contribute to the pathogenesis of DR.
