摘要
乳杆菌能够维持肠道内正常菌群的平衡,而来源于动物肠道的乳杆菌更容易在肠道内生长繁殖,所以分离冷水鱼肠道内的低温乳杆菌具有重要的应用价值。利用MRS、M17、改良番茄和乳杆菌选择性琼脂培养基4种培养基,从6种不同的冷水鱼肠道以及粘膜中分离筛选出耐低温的乳杆菌20株。通过一些生理生化试验初步确定疑似乳杆菌菌株24株,并测定其最适生长温度,其中有7株为嗜冷菌,17株为耐冷菌。根据16Sr RNA基因序列构建系统发育树可知,20株菌隶属Lactobacillius spp.,其他4株分别隶属Clostridium spp.和Pseudomoncs spp.。
Lactobacillus can balance the normal flora in the intestines,and the Lactobacillus from animal intestinal are more easily to grow in the intestinal tract.So screening of Lactobacillus from cold water fish intestinal tract has an important application value.There are 20 Lactobacillus strains which were isolated from 6 intestinal and mucosa of cold water fishes.These Lactobacillus were isolated by four different culture medium for MRS,M17,tomato juice agar (modified) and Lactobacillus selective agar.We determined 24 strains suspected Lactobacillius strains with using some normal physiological and biochemical experiments,then determined the optimum growth temperature.During the 24 suspected Lactobacillius strains,there are 7 strains for the psychrophilic and 17 strains for the psychrotolerants.According to the 16S rRNA gene sequences to construct phylogenetic tree,it was found that 20 isolates belonged to Lactobacillius spp.,and the rest of the 4 strains belonged to Clostridium spp.and Pseudomoncs spp..
