摘要
目的 探讨用白细胞介素 2 (IL - 2 )激活的外周血 (APB)、脐带血 (ACB)和骨髓 (ABM)单个核细胞的培养上清对肿瘤细胞的作用 ,及其作用的可能机制。方法 应用 MTT法和集落培养法在体外检测了 APB、ACB和ABM培养上清对 HL 6 0和 K5 6 2细胞的作用 ,用 EL ISA法检测了各种上清中 TNF-α和 IFN-γ的产生情况。结果 APB、ACB和 ABM培养上清都可以产生 TNF-α和 IFN-γ,对 HL 6 0和 K 5 6 2细胞有一定抑制作用 ,以 APB培养上清最强。结论 应用 IL - 2净化自体干细胞移植物中混杂的肿瘤细胞时 ,培养上清中的细胞因子或其它成分也可以直接抑制肿瘤细胞。
Objective To investigate the effect of the supernatants of interleukin 2 activated peripheral blood (APB),cord blood (ACB),and bone marrow (ABM) mononuclear cells on the leukemia cell lines,and its mechanism.Methods MTT and colony culture methods were used to detect the effects of the supernatants of APB,ABM and ACB on the HL60 and K562 cells,at the same time the concentration of TNF α and IFN γ in those supernatants wer measured by ELISA method.Results Supernatants of APB,ACB and ABM suppressed the proliferation of the tumor cell lines K562 and HL60.TNF α and IFN γ were detected in those supernatants.Conclusion The cytokines or other components in the culture supernatants may directly suppress the tumor cells.
