Determination of Nisoldipine Concentration in Human Plasma by Capillary Gas Chromatography Using Electron-Capture Detection

摘要 目的 :建立测定人血浆中尼索地平的毛细管气相色谱电子捕获检测法。方法 :色谱柱为 2 5m× 0 32 8mmDB- 1交联熔融毛细管柱 ,膜厚 0 5 2 μm ,检测器为63Ni电子捕获检测器。内标为尼莫地平 ,血浆样品在碱性条件下用乙醚 -环己烷 (1∶1)提取。结果 :在 0 1~ 30ng·mL-1浓度范围内峰面积比与浓度呈良好的线性关系 ,r =0 9997。人血浆中尼索地平的最低检出浓度为 5 0pg·mL-1。高、中、低 3种浓度的平均回收率为 91 92 %~ 97 89% ,日内和日间精密度的RSD分别小于 4 6 %和 6 9%。结论 Objective:To establish a capillary GC-ECD method for determining nisoldipine in human plasma.Methods:Chromatography was performed on a 25 m×0 328 mm (0 52 μm film thickness),DB-1 fused-silica capillary column coupled with a 63 Ni electron-capture detector.The internal standard was nimodipine.After making alkaline with 1 mol·L -1 NaOH solution,the plasma was extracted with ethylether:n-hexane(1∶1).Results:A good linearity was obtained from 0 1 ng·mL -1 to 30 ng·mL -1 of nisoldipine in human plasma with a correlation coefficient of 0 999 7 The detection limit of nisoldipine in human plasma was 50 pg·mL -1 .The extraction recovery was 91 92%~97 89%.The RSD values of intraday and interday were lower than 4 6% and 6 9% respectively.Conclusion:The established method was provided to be a reliable quantitative method for pharmacokinetic studies of nisoldipine.
