摘要
建立了 L C- MS法测定人血浆中尼索地平的浓度。血浆在碱性条件下用乙醚 -正己烷 (1∶ 1)提取 ,以尼莫地平为内标 ,采用 Inertsil C1 8色谱柱 ,AP- ESI质谱检测器 ,正离子模式 ,采集方式为 SIM。流动相为甲醇 -水 -醋酸 (75∶2 5∶ 0 .1)。尼索地平在 0 .2~ 4 0 ng/ ml范围内线性关系良好 ,提取回收率大于 80 % ,RSD小于 10 % ,定量限为0 .2 ng/ m
A LC-MS method for determination of nisoldipine in human plasma was presemed. The basified plasma was extracted by ether-n-hexane(1∶1), and nimodipine was used as the internal standard. An Inertsil C 18 column was used with an electrospray ionization mass spectrometer in positive ion and selected-ion monitoring mode. The mobile phase was methanol-water-acetic acid(75∶25∶0.1).The calibration curve was linear in the concentration range of 0.2~40 ng/ml. The extraction recovery was more than 80%, and RSD was less than 10%. The detection limit of nisoldipine was 0.2 ng/ml.
