摘要
目的 :探讨阿霉素诱导细胞凋亡的分子机制。方法 :通过光镜和电镜观察阿霉素作用不同时间后的形态学改变 ,DNA琼脂糖凝胶电泳、流式细胞仪分析 DNA含量和细胞周期 ,并检测细胞 p5 3基因蛋白表达 ;免疫组织化学法检测 bcl- 2基因蛋白的表达。结果 :阿霉素作用于 MEC- 1细胞后出现了细胞凋亡的特征 ,p5 3蛋白表达的荧光指数 (FI)值分别为 0 .2 0± 0 .0 4、0 .42± 0 .11、1.6 7± 0 .17,bcl- 2基因蛋白表达逐步降低。结论 :p5 3基因和bcl- 2基因参与凋亡过程可能是阿霉素诱导 m Ec-
Objective:To explore whether or not adriamycin inhibits mEc cells by apoptosis. Methods:Adriamycin was used to treat mucoepidermoid carcinoma cells mEc 1. The morphologic changes were observed at different times by light and electronic microscope. DNA fragments were shown on agarose gel electrophoresis. DNA content and cell cycles were analyzed by flow cytometry. The changes of p53 gene was monitored by flow cytometer, bcl 2 gene was determined by immunohistochemical method.Results:The results showed that MEC 1 cells presented classic morphlogic features of apoptosis, FI expression of p53 gene protein was 0.20±0.04, 0.42±0.11, 1.67±0.17 after cell apoptosis induced by adriamycin,and expression of bcl 2 protein was decreased.Conclusion:The apoptosis followed by the changes of p53 and bcl 2 in human salivary gland mucoepidermoid carcinoma cells is the main mechanism of adriamycin inhibited tumor cell growth.
