摘要
【目的】探讨低分子肝素(LMWH)对裸鼠胃癌原位模型生长、转移及趋化因子SDF-1表达的影响。【方法】应用原位移植方法建立裸鼠胃癌原位模型,术后7d将裸鼠随机分为3组:对照组(生理盐水)、化疗组(5-氟尿嘧啶)、LMWH组(低分子肝素钙),分别每周注药2次,共5周,第6周处死动物,观察裸鼠胃癌原住模型肿瘤转移及生长情况。采用RT—PCR技术检测胃肿瘤组织SDF-1mRNA表达水平。【结果】化疗组、LMWH组肿瘤体积较对照组明显缩小,差异有统计学意义(P〈0.05);LMWH组胃癌原位模型肿瘤转移率(20%)低于对照组(70%),差异有统计学意义(P〈0.05),而与化疗组之间差异无统计学意义(P〉o.05);LMWH组SDF-1mRNA表达均较对照组和化疗组低,差异有统计学意义(P〈0.05)。【结论】LM—wH可抑制裸鼠胃癌原位模型的肿瘤生长及远处转移,其部分机制可能与降低肿瘤sDF_1的表达有关。
[Objective] To explore the effect of low-molecular weight heparin(LMWH) on tumor metasta- sis and growth and the expression of SDF-1 in orthotopic transplantation gastric tumor model of nude mice. [Methods] The models of orthotopic transplantation gastric cancer in nude mice was established by orthotopic transplantation. All nude mice were randomly divided into control group(normal saline), chemotherapy group (5-fluorouracil) and LWMH group(LWMH). The treatment duration was twice a week for 5 weeks. The animals were sacrificed at 6 weeks. The tumor growth and metastasis of orthotopic gastric cancer in nude mice were observed. RT-PCR was used to detect the expression of SDF-1 mRNA in gastric cancer tissues. [Re sults] Compared with control group, the tumor size in chemotherapy group and LMWH group were reduced obviously, and there was significant difference( P 〈0.05). The tumor metastasis rate in LMWH group was lower than that in control group(20% vs. 70%), and there was significant difference between LMWH group and control group( P 〈0.05), but there was no difference between LMWH group and chemotherapy group( P 〉0.05). The expression of SDF-1 mRNA in LMWH group was lower than that in control group and chemo therapy group, and there was significant difference( P 〈0.05). [Conclusion] LMWH can inhibit the tumor growth and distant metastasis of orthotopic transplantation gastric cancer in nude mice. The mechanism may be associated with the decreasing of SDF-1 expression.
