齐墩果酸对白血病HL-60细胞PI3K、Akt、Cav-1表达的影响 被引量:4

摘要 目的:研究齐墩果酸(Oleanic acid,OA)对人白血病HL-60细胞PI3K、Akt、Cav-1表达的影响。方法:OA作用HL-60细胞后,采用透射电镜观察细胞形态,RT-PCR法检测HL-60细胞PI3K、Akt、Cav-1的mRNA表达。结果:OA作用于HL-60细胞后,细胞形态改变明显,出现凋亡小体,且PI3K、Akt、Cav-1的mRNA表达下调(P<0.01)。结论:OA可抑制HL-60细胞增殖,这可能与其下调HL-60细胞中PI3K、Akt、Cav-1的表达有关。 Objective:To study the effect of oleanolic acid (OA) on the expression of pi3k,akt and cav - 1 genes in leukaem HL -60 cells. Methods:In HL -60 cells, after being treated by OA, the morphological and structural changes of the cells were observed by transmission electro microscopy; the mRNA expressions of pi3k,akt and car - 1 were measured by RT - PCR. Results: After being treated by OA, the HL - 60 cells had obvious mor- phological and structural changes under the 'transmission electro microscopy, and apoptosis body was observed as well. The mRNA expression of pi3k and akt were down regulated (P 〈 0. 01 ), the mRNA expression of cav - lwas up regulated (P 〈 0. 01 ). Conclusion: OA could inhibit the proliferation of HL - 60 cells by down - regulating the expression of PI3 K and Akt , by up - regulating the expression of CAV - 1.
