摘要 PI3K/Akt/mTOR信号途径作为细胞内重要信号转导通路之一,在维持细胞增殖、存活和凋亡中发挥关键作用。当PI3K/Akt/mTOR通路分子的基因功能失常、上游调控信号异常放大时使该信号通路异常激活,细胞生存与凋亡失衡,正常细胞发生恶性转化。研究表明,PI3K/Akt/mTOR信号通路在急性早幼粒细胞白血病存在异常激活,对肿瘤细胞的生存、增殖、分化、凋亡和耐药性中起重要作用。现就近年来在PI3K/Akt/mTOR信号通路与急性早幼粒细胞白血病关系研究领域的进展予以简要综述。 Phosphatidylinositol 3-kinase(PI3K)/Akt/mammalian target of rapamycin(mTOR) signaling axis is an important intracellular signal transduction pathway and plays a central role in cell proliferation,growth,and survival under physiological conditions.Aberrant activation of PI3K/Akt/mTOR signaling due to genetic dysfunction and abnormal magnification of upstream regulatory signal results in the imbalance between cell viability and apoptosis and malignant transformation.It has been shown that the constitutive activation of PI3K/Akt/mTOR signaling network in acute promyelocytic leukemia plays a vital role in survival,proliferation,differentiation,apoptosis,and drug-resistance of leukemic cells.This article reviews recent progress on PI3K/Akt/mTOR signaling axis in acute promyelocytic leukemia.
