摘要
目的:观察益气化浊胶囊对KKAy小鼠肝脏中胰岛素受体底物1(IRS-1)和2(IRS-2)mRNA和蛋白表达的影响,探讨其增加胰岛素敏感性的作用机制。方法:KKAy小鼠随机分为模型组、益气化浊高剂量组、低剂量组和阳性组,C57BL/6J小鼠作为正常组,每组8只。给药8周后,测空腹血糖(FPG)、血清胰岛素(FINS)及胰岛素抵抗指数(HOMA-IR),采用Real-time PCR技术和Western blot印迹法检测肝脏中IRS-1、IRS-2 mRNA及蛋白表达水平。结果:与模型组比较,高剂量组FPG、FINS、HOMA-IR降低,IRS-1、IRS-2mRNA和蛋白表达水平升高;低剂量组FPG降低,IRS-2蛋白表达明显升高。结论:益气化浊胶囊改善KKAy小鼠胰岛素抵抗,可能与上调肝脏中IRS-1、IRS-2 mRNA和蛋白表达水平有关。
Objective :To observe effects of Yiqi Huazhuo Capsule on expression of insulin receptor substrate 1 (IRS-1) and IRS-2 mRNA and proteins in liver to explore its possible molecular mechanism in increasing insulin sensitivity. Methods :KKAy mice were randomly divided into model group,Yiqi Huazhuo Capsule of high dose group, low dose group, positive control group, and C57BL/6J mice were normal control group, which group had 8 mice. 8 weeks after administration, measuring fasting blood glucose (FBG), fasting insulin (FINS) and calculated insulin resistance index (HOMA-IR). The expressions of IRS-1 and IRS-2mRNA and proteins were detected with real-time fluorescence quantitative PCR method and Western blot method. Results: As compared with model group, FPG,FINS,HOMA-IR were decreased and mRNA and protein expression levels of the IRS-1 and IRS-2 were increased in high group, FPG was decreased and protein expression levels of IRS-2 was increased in low group. Conclusion:Yiqi Huazhuo Capsule improves FPG and insulin resistance in KKAy mice and its mechanism is probable related to raising expression level of IRS-1 and IRS-2 mRNA and proteins in liver.
