摘要
利用超声波振断法将水稻细菌人工染色体 (BAC)基因组文库———Nipponbare库中的一个BAC克隆 (E5 0 )振断 ,产生许多大小不同的DNA随机片段 ,回收其中 1 7~ 3 5kb的片段 ,并将这些随机片段克隆入质粒载体pUC18。
For sequencing the whole BAC by shotgun, a subclone library for the rice megabase DNA, a BAC from the Nipponbare library that was a bacterial artificial chromosome library of rice genome, was constructed. The BAC was sonicated and destroyed to small DNA fragments. The 1 7~3 5kb DNA fragments were extracted and cloned to pUC18.
