摘要
目的研究游泳运动、二甲双胍及两者联合干预对肥胖大鼠附睾脂肪、皮下脂肪、肌肉组织chemerin蛋白表达的影响,探讨其与机体的内在联系。方法将70只雄性SD大鼠随机分为普通饲料喂养组(NC组,n=10)和高脂饲料喂养组(HD组,n=60)。16周后高脂饮食诱导的肥胖大鼠建模成功(n=40),随机分为4组:高脂饲养对照组(HF组,n=10)、游泳运动干预组(SWI组,n=10)、二甲双胍干预组(MET组,n=10)、游泳运动+二甲双胍联合干预组(SAM组,n=10)。利用Western blot法检测大鼠附睾脂肪、皮下脂肪和肌肉组织各组别chemerin蛋白表达水平。结果①高脂饲养22周后,HF组附睾脂肪、皮下脂肪及肌肉组织的chemerin蛋白表达较NC组增加(P<0.05);与皮下脂肪及肌肉组织相比,附睾脂肪的chemerin蛋白表达增加更为显著(P<0.05);②经过干预后,SWI组、MET组、SAM组附睾脂肪和皮下脂肪chemerin蛋白表达较HF组降低(P<0.05);与SWI组和MET组相比,SAM组的chemerin蛋白表达降低更为显著(P<0.05);在肌肉组织中,无论干预前后,SWI组、MET组、SAM组与HF组chemerin蛋白表达差异均无统计学意义(P>0.05)。结论①正常SD大鼠高脂饲养后可引起肥胖及脂肪和肌肉组织中chemerin蛋白表达增加;②游泳运动和二甲双胍及两者联合干预均可显著降低脂肪组织,尤其是内脏脂肪组织中chemerin蛋白的表达。
Objective To observe the effects of swimming exercise, metformin and the combination of both interventions on chemerin protein expression levels in epididymis adipose tissue (EAT) , subcutaneus adipose tissue (SAT) and muscle tissue (MT) of obesity rats and to explore the relationship between chemerin and obesity-related metabolic disorders. Methods The 70 male conventional SD rats were divided into control diet group ( n = 10) and high fat group (n = 60). After 16 weeks obesity model( n = 40) was established and was randomly divided into four groups : high fat group (HF) , swimming group (SWI) , metformin group (MET) , and the combination of swimming and metformin group(SAM) with 10 rats in each group. Chemerin protein expression levels in EAT, SAT and MT were determined by Western blot. Results ① After 22 weeks high fat diet induced obesity, chemerin protein expression levels of obesity group in EAT, SAT and MT, especially that of EAT increased compared to NC group(P 〈 0.05). ② After intervention, in EAT and SAT, chemerin expression levels of the group of SWI, MET and SAM significantly decreased, especially SAM. Chemerin expression levels of the group of MET, SWI and SAM in MT decreased with no statistical difference. Conclusion ① High fat diet could induce SD rats obesity and also increase the expression levels of chemerin protein in adipose tissue and muscle tissue. ② Swimming, metformin and the combination of both intervention could down-regulate the level of chemerin, especially in visceral adipose tissue.
