摘要
受无糖培养技术启示,在太子参组培苗育苗过程中,以液体培养基取代固体培养基,应用纸桥法,以太子参幼苗茎段为外植体,经有效诱导芽分化后转入以蛭石为基质的增殖培养基增殖培养,并诱导生根,成苗后辅以自然光照,通过培养基改进对太子参组培苗质量进行研究。结果表明:茎段在合MS+0.1mg/LNAA+0.5~1.0mg/L6-BA+2%蔗糖的液体培养基滤纸桥上的芽分化率达80%以上。不定芽在以蛭石为基质的MS+0.1mg/LNAA+1mg/L6.BA十2%蔗糖的培养基中增殖并生根,生根后的试管苗转到低糖(0.5%)或无糖培养基中,经自然光照培养能得到高质量的试管苗。
Inspiraed by sugar-free culture technique, liquid culture medium replacement for solid culture medi- um, the stems of pseudostellaria heterophylla have been used as explants which incubate on filter paper bridg- es in test tubes containing liquid basal medium to study quality in tissue-cultured shoots. The results show that MS+0. lmg/L NAA+0.5 ~ 1.0mg/L 6-BA+2% sucrose was liquid basal medium for filter paper bridges, and the shoot differentiation rate was up to 80%. The adventitous sprouts are propagated and rooted in ver- miculite medium of MS+0. 1 mg/L NAA+ 1mg/L 6-BA+2% sucrose. Then high quality test-tube plantlets were cultivated in low sugar (0.5%) or sugar-free culture medium by natural lighting.
