摘要
研究了以濒危植物珙桐叶片作为外植体诱导愈伤组织及分化再生植株.结果表明:本实验中,诱导愈伤组织的最佳培养基为1/2MS+2.0 mg/L IBA+1.0 mg/L 6-BA;愈伤组织继代的最佳培养基为1/2MS+3.0 mg/L IBA+1.5 mg/L 6-BA最佳分化培养基为MS+2.0mg/L 6-BA+1.0 mg/L IBA+0.2 mg/L GA_3,芽分化率为6.2%;生根最佳培养基为1/2MS+0.4 mg/L IBA,生根率为60%.
A protocol for dedifferentiation and plantlet regeneration of Davidia involucrate, a rare and endangered species but very important for breeding purposes had been investigated through in vito leaves culture. The results showed: in this experiment, the best callus induction medium was 1/2MS+2.0 mg/L IBA+1.0 mg/L 6-BA; the best callus proliferation medium was 1/2MS+3.0 mg/L IBA+1.5 mg/L 6-BA; the highest adventitious shoot regeneration is 6.2% of the explants with the medium of MS +2.0 mg/L 6-BA+1.0 mg/L IBA+-0.2 mg/L GA3+a high rooting frequency(60%) was recorded on medium of 1/2MS+0.4 mg/L IBA.
