摘要
目的探讨环氧合酶-1(cyclooxygenase,Cox-1)在慢性非细菌性前列腺炎(CNP)疼痛中的作用及可能机制。方法wistar大鼠60只,随机分为0、2、4、6、8w制备抗原组,利用免疫组织化学法建立慢性非细菌性前列腺炎模型。通过行为学观察、病理组织学检查、免疫组织化学SABC法检测Cox-1在前列腺及脊髓中的表达变化。结果造模后大鼠活动减少,弓背向上,被毛竖起,舔、咬非注射部位,出现逃避行为,对抓捕反应敏感。造模后2w大鼠前列腺腺体规则,问质开始出现少量淋巴细胞浸润,随时间延长炎症表现加重,直至6w和8w前列腺部分腺上皮脱落,问质出现大量片状淋巴细胞、间质纤维化,呈现显著炎症病理改变。Cox-1在各组前列腺中均有阳性表达,沿腺泡内衬上皮呈连续性分布,间质不表达。Cox-1在各组前列腺中的表达差异均无统计学意义(P〉0.05)。Cox-1在各组大鼠脊髓L5~s2节段后角中均有表达,阳性细胞分布在脊髓浅层,表达于神经元胞浆。与0w组相比,2w组表达无变化,差异无统计学意义(P〉O.05),4w组、6w组、8w组阳性只数增加,差异有统计学意义(P〈0.05);与2w组相比,6w组、8w组阳性只数增加,差异有统计学意义(P〈O.05)。结论Cox-1对慢性非细菌性前列腺炎疼痛的形成和维持起重要作用。
Objective To explore the role and mechanism of Cyclooxygenase-1 in Chronic nonbacterial pros tatitis. Methods 60 Wistar rats were randomly divided into 0 week group, 2 week group, 4 week group, 6 week group, 8 week group and antigen group and the chronic nonbacterial prostatitis model was built by adopting immunohistochemistry. The ethological and pathological changes of rats were observed and the expression of Cox-1 by immunohistochemistry was detected. Results The activity of rats was reduced, the pain hypersensitivity was showed and the capture reaction became too sensitive in rats molding. The pros- tate gland was normal but a small amount of lymphocyte began to appear interstitial after 2 weeks building. The significant inflammation were presented in the scope of lesions, lymphocyte infiltration and inter- stitial hyperplasia until 6 weeks. Compared with 0 weeks group, the expression of 2 weeks, 4 weeks, 6 weeks, 8 weeks group in the prostate had no change, there was no statistically significant difference (P 0.05) ; Compared with group 2 weeks, the expression of 4 weeks, 6 weeks, 8 weeks group had no change, there was no statistically significant difference; Compared with group 4 weeks, the expression of 6 weeks, 8 weeks group had no change, there was no statistically significant difference; the expression of 6 weeks and 8 weeks group had no change, there was no statistically significant difference. Compared with 0 weeks group, the expression of group 2 weeks had no change in the spinal cord, there was no statistically signifi- cant difference (P 〉 ses, the difference w weeks, 8 weeks group sion: Cox-1 played an 0.05), the expression of 4 weeks, 6 weeks, 8 weeks group had increased positive ca as statistically significant (P 〈0.05) ; Compared with group 2 weeks, the expression of 6 had increased positive cases, the difference was statistically significant (P 〈0.05). C.onelu- important role in the occurrence of pain in chronic nonbacterial prostatitis.
