摘要
目的:用雄性Wistar大鼠前列腺组织抗原蛋白诱导自身免疫性前列腺炎动物模型。方法:取5只雄性大鼠,在无菌条件下取出前列腺组织,按双缩脲法测定蛋白含量,以生理盐水稀释至40mg/mL,再以完全弗氏佐剂倍量乳化。将正常30只动物分为5组,每组6只。除正常组外,每组动物腹侧皮下多点注射乳化后的抗原液1mL,分别于2、4、6、8周组取前列腺组织,在光学显微镜下观察前列腺组织病理形态。结果:造模2周后,前列腺各腺管之间可见散在的淋巴细胞,造模8周后,前列腺间质及腺体中可见大量的弥散的淋巴细胞浸润。结论:应用大鼠前列腺组织抗原蛋白诱导出自身免疫性前列腺炎动物模型,为前列腺炎的自身免疫性病因学说提供了实验依据。
Objective To establish experimental autoimmune prostatitis animal model in Wistar rats using the rats' Prostate antigen. Methods Five Wistar rats' prostatic tissue was taken out under germfree conditions. After thc protein levels were measured by the biuret method, the protein concentration was diluted into 40 mg/mL and double emulsified by Freund's complete adjuvant(FCA). Thirty rats were divided into five gToups with 6 rats each. In the 4 tested groups the control group, 1 mL emulsified antigen was injected into each rat subcutaneously at abdominal wall multi - loci. The prostate tissues were taken out at 2,4,6,8 weeks after the injection respectively, and observed under the light microscopy. Result Two weeks after the injection, the lymphocytes were interspersed among the glandular tube. Eight weeks after injection, plenty of lymphocytes were observed among the interstitial tissue and the glands. Conclusion This autoimmune prostatitis animal model induced by subcutaneous injection of the rats' s prostate protein provided an experimental base for the autoimmune theory of prostatitis.
