新诊断2型糖尿病患者外周血单个核细胞蛋白酪氨酸磷酸酶1B的表达 被引量:1

Expression of protein-tyrosine phosphatase 1B mRNA of peripheral blood mononuclear cells in newly diagnosed type 2 diabetic patients

摘要 目的探讨蛋白酪氨酸磷酸酶1B(protein tyrosine phosphatase 1B,PTP1B)在新诊断的2型糖尿病患者外周血单个核细胞(peripheral blood mononuclear cell,PBMC)中的表达水平及意义。方法采用实时荧光定量PCR法检测20例新诊断2型糖尿病患者(糖尿病组)和20例血糖正常者(对照组)PBMC中PTP1BmRNA的表达,分析其与体质量指数(body mass index,BMI)、空腹血糖(fasting plasma glucose,FPG)、空腹胰岛素(fasting insulin,FINS)、稳态模型的胰岛素抵抗指数(homeostasis model assessment of insulin resistance,HOMA-IR)、血脂的相关性。结果糖尿病组PBMC中PTP1BmRNA水平(1.05±0.19)明显高于对照组(0.53±0.17)(P<0.01),且PTP1BmRNA表达与BMI、FPG、FINS、HOMA-IR、三酰甘油和总胆固醇呈正相关(r=0.58,r=0.78,r=0.55,r=0.73,r=0.68,r=0.59,P均<0.01)。结论 PTP1B在新诊断2型糖尿病患者PBMC中表达较血糖正常者增加,其高表达与新诊断2型糖尿病胰岛素抵抗相关。 Objective To explore the expression level of protein tyrosine phosphatase 1B (PTPIB) mRNA of peripheral blood mononuclear cells (PBMCs) in newly diagnosed type 2 diabetic patients. Methods The expression of PTPIB mRNA in PBMC was detected by real-time quantitative PCR in 20 cases of newly diagnosed type 2 diabetic mellitus (T2DM group) and 20 controls with normal plasma glucose (control group). The correlation of PTPIB mRNA with body mass index (BMI), fasting plasma glucose (FPG), fasting insulin (FINS), homeostasis model assessment of insulin resistance (HOMA IR) and lipid were analyzed. Results The PTPIB mRNA level of PBMC was significantly higher in T2DM group (1.05=0.19) than that in control group (0.53±0.17) (P〈0.01). The expression of PTP1B mRNA was positively correlated with BMI, FPG, FINS, HOMA-IR, triaeylglycerol and total cholesterol (r=0. 58, r=0. 78, r= 0.55, r=0.73, r=0. 68, r=0. 59,P%0.01). Conclusion The expression of PTPIB mRNA in PBMC is higher in newly diagnosed type 2 diabetic patients than that in those with normal plasma glucose, the high expression of which is correlated with insulin resistance.
