杧果内生真菌分离鉴定及多样性分析 被引量:10

摘要 【目的】为了解杧果内生真菌的群落组成、物种多样性及分布规律,【方法】采用形态学和ITS-rDNA序列分析相结合的方法对‘凯特’等4个杧果品种的内生真菌进行了分类鉴定,并以内生真菌的分离率(IR)、定殖率(CR)、相对频率(RF)、多样性指数(H')及相似性系数(Cs)为指标衡量试验结果。【结果】从480块新鲜杧果组织中分离得到53株内生真菌,经鉴定为16个分类单元,其中14个属于子囊菌(Ascomycota),2个属于担子菌(Basidiomycota)。胶孢炭疽菌(Colletotrichum gloeosporioides)、杧果球座菌(Guignardia mangiferae)、葡萄座腔菌(Botryosphaeria dothidea)为杧果的优势种,相对频率分别为20.75%、18.87%和18.87%。【结论】杧果内生真菌种类丰富,且具有品种和组织差异性。 [Objective ]The objective of the study were to analyze the composition, diversity and distribu tion of endophytic fungal community of Mangifera indica Linn(Mango). [Method]The endophytic fungi iso lated from the four kinds of mango were identified and classifed by the morphological observation and ITS sequence analysis. The result were analyzed by the indexes such as isolation rate (IR), colonization rate(CR), relative frequency(RF), ShannonWiener biodiversity index (H') and similarity coefficient (Cs). [Result]Fifty three strains of endophytic fungi were isolated from the 480 pieces of mango organizations. They were classified and identified to 16 taxa, 14 species of Ascomycota and 2 species of Basidiomycota were obtained. The dominant groups of those endophytic strains were Colletotrichum gloeosporioides, Guignardia mangiferae and Botryosphaeria dothidea, accounted for 20.75%, 18.87% and 18.87%, re spectively.[Conclusion]The composition of endophytic ftmgal community in mango has a certain diversity, and the distribution of endophytic fungi has the species and tissue specificity.
