摘要
目的:使用2次小剂量注射链脲佐菌素(STZ)建立大鼠糖尿病肾病(DN)模型,接着服用苦寒中药煎剂制造脾肾阳虚证。方法:采用高糖高脂饲料加两次小剂量腹腔注射STZ的方法诱导糖尿病肾病模型。再将所有造模成功大鼠予腺嘌呤及苦寒中药(龙胆草、栀子、番泻叶)混合灌服,完成糖尿病肾病脾肾阳虚型大鼠模型。观察造模后动物体重、饮食、体温、精神等,以评价动物模型脾肾阳虚存在的指标。大鼠随机分为模型组、正常组。进行血清FBG、SCr、24 h尿蛋白定量、肾重指数测定及肾小球病理学分析。结果:与空白组比较,模型组肾脏病理切片有明显的肾小球硬化;FBG、SCr、24 h尿蛋白定量、ET、肾脏指数均升高(P≤0.05)。症状大鼠明显消瘦,多饮、多尿,精神萎靡,倦怠,蜷缩,反应迟钝,动作迟缓,毛发无光泽。结论:该动物模型是一个更符合临床糖尿病肾病脾肾阳虚证发病过程的动物模型,为脾肾阳虚的证型实质探讨提供了较为理想的动物模型。
Objective:To establish the rat model with diabetic nephropathy (DN) through twice small dose injection of streptozotocin (STZ), then using the bitter and cold traditional Chinese medicine decoction to sicken the rats with spleen - kidney yang deficiency syndrome. Method:We used the high fat, high sucrose diet and twice small STZ injection (i. p. ) to establish the rat model with DN. And after intragastrically administering the successful model with adenine and bitter - cold traditional Chinese medicine (gentian, cape jasmine, folium sennae), we got the DN spleen -kidney yang deficien- cy rat model. Results : Compared with the blank group, kidney pathological section of the model rats showed significant glo- merular sclerosis, and increased FBG, SCr, 24 hour urine protein, ET and kidney index ( P ≤ 0.05 ). These rats also seemed to be emaciated, polydipsia, potyuria, sluggish, lassitude and showed a drooping mood, retard mind and behav- ior and dull hair. Conclusion : This rat model is more in line with the clinically progress of DN spleen - kidney yang defi- ciency, and thus better serves as the animal model to discuss the essence of spleen - kidney deficiency syndrome.
