正交试验法分析平帕汤治疗帕金森病的最佳配伍比例 被引量:5

An Analysis of Compatibility Proportions of Pingpa Decoction for Parkinson's Disease with Orthogonal Test Method

摘要 目的探讨平帕汤治疗帕金森病(PD)的配伍规律并确定其最佳配伍比例。方法112只C57/BL雄性小鼠,采用1-甲基-4-苯基-1,2,3,6-四氢吡啶(MPTP)腹腔注射法建立PD模型,随机分为中药1~8组,每组14只。按L8(2^7)正交设计表,各组以相应配伍比例的平帕汤(银杏叶、肉苁蓉、罗布麻、山梗菜)灌胃,每日1次,给药10天后,采用高效液相电化学法(HPLC-ECD)检测各组小鼠纹状体内多巴胺(DA)、3,4-二羟基苯乙酸(DOPAC)和高香草酸(HVA)含量;采用免疫组化染色法观察各组小鼠黑质酪氨酸羟化酶(TH)阳性细胞点光密度(IOD值)。结果各组小鼠黑质致密部可见阳性神经元呈条带状,胞体呈梭形或锥形,胞浆深棕黄色到浅黄色不等,可见轴突纤维同时伴有不同程度上的胞间纤维缺失。平帕汤治疗PD的影响因素主次顺序为银杏叶〉肉苁蓉〉山梗菜〉罗布麻,银杏叶、肉苁蓉及二者的交互作用为主要影响因素,其中银杏叶及其与肉苁蓉交互作用的差异有统计学意义(P〈0.05)。结论平帕汤最佳配伍比例为银杏叶20g、肉苁蓉40g、罗布麻20g、山梗菜20g。 Objective To study the compatibility law and optimal compatibility proportion of Pingpa Decoction for Parkinson's disease (PD). Methods Totally 112 C57/BL male mice were induced into PD models with intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and randomized into 8 Chinese medication groups, with 14 in each. According to the L8 (2^7) orthogonal array, each group was gavaged with corresponding Pingpa Decoction [Yinxingye ( Foliurn Ginkgo ) , Roucongrong ( Herba Cistanches ) , Luobuma (Folium A poc yni Veneti ) and Shangengcai (Lobelia sessili f o- lia Lamb)] once daily for 10 consecutive days. The dosage in Chinese medication group 1, group 2 to 7 and group 8 was 12g/ (kg· d), 24g/(kg · d) and 48g/(kg · d) respectively. High performance liquid chromatography electrochemical detector (HPLC-ECD) was used to determine the contents of dopamine (DA), 3, 4-dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA) in striatum. Immunohistochemical method was used to observe the expression of tyrosine hydroxylase (TH) in the substantia nigra. Results Banded positive neurons, spindle or tapered cell body, dark brown yellow to pale yellow cytoplasm, axon fibers and missing intercellular fibers could be seen in the substantia nigra of mice in each group. The therapeutic effect of Pingpa Decoction for PD was influenced by Yinxingye, Roueongrong, Shangengcai and Luobuma in order, especially Yinxingye and the interaction of Yinxingye and Roueongrong (P〈 0.05). Conclusion The optimal compatibility proportion of Pingpa Decoction is 20g Yinxingye, 40g Roucongrong, 20g Luobuma and 20g Shangengeai.
