摘要
目的:研究血清中miR-29a-5p和miR-222表达与肝细胞肝癌(HCC)的诊断价值和预后的关系。方法:采用实时荧光定量PCR(RT-PCR)对比检测76例HCC患者、62例肝良性疾病患者和60例健康对照组血清中miR-29a-5p和miR-222水平。分析miR-29a-5p和miR-222的表达与患者临床病理参数以及肝癌切除术预后的关系。结果:HCC患者血清miR-29a-5p和miR-222表达明显高于良性肝病和正常对照组(P<0.05),与AFP、肿瘤大小、癌栓、病理分化、TNM分级有关(P<0.05)。HCC患者中miR-29a-5p和miR-222低表达组的复发转移率显著低于高表达组,其术后生存率也显著高于高表达组(P<0.05)。结论:血清miR-29a-5p和miR-222与HCC的临床病理特征密切相关,不仅是潜在的HCC早期检测指标,且miR-29a-5p和miR-222在HCC患者血清中表达上调与HCC复发转移率高及预后差密切相关,提示其也可能是判断HCC手术预后的分子标志物。
Objective：To investigate the clinical significance of serum miR-29a-5p and miR-222 expression in the diagnosis and prognostic evaluation of hepatocellular carcinoma（HCC）.Methods：The serum samples were collected from 76 patients with HCC,62 patients with benign disease and 60 normal subjects.The expression levels of miR-29a-5p and miR-222 in the serum were measured by reverse transcription polymerase chain reaction （RT-PCR）.The correlations of miR-29a-5p and miR-222 expression to the clinicopathological features and prognosis were analyzed.Results：HCC patients had significantly higher serum miR-29a-5p and miR-222 levels than benign diseases and healthy controls.The expression level of serum miR-29a-5p and miR-222 had a close correlation with AFP level,tumor size,tumor thrombi,histological differentiation,TNM classification （P ＜ 0.05）.The survival time of HCC patients with a lower serum miR-29a-5p and miR-222 expression levels was longer than those with a higher level,and the recurrent/metastatic of HCC patients with a higher serum miR-29a-5p and miR-222expression levels was also higher than those with lower expression （P ＜ 0.05）.Conclusion：The expression of miR-29a-5p and miR-222 in serum had close correlations with the clinicopathologic features of HCC,and the up-regulated expression of serum miR-29a-5p and miR-222 is corelated with postoperative metastasis and invasion and poor prognosis,indicating that miR-29a-5p and miR-222 can not only be used as a potential marker for HCC,but also as a prognostic marker in HCC patients.
