脐带间充质干细胞联合番茄红素的抗衰老作用 被引量:2

摘要 背景:研究证实脐带间充质干细胞、番茄红素均有不同程度的抗衰老作用,而关于二者联合抗衰老作用的研究国内外研究甚少,目前基本处于空白状态。目的:观察人脐带间充质干细胞联合番茄红素对D-半乳糖衰老模型小鼠的抗衰老作用。方法:实验选用ICR小鼠,通过颈背部皮下注射D-半乳糖复制亚急性衰老小鼠模型,分为对照组、模型组、干细胞治疗组、番茄红素治疗组和干细胞联合番茄红素治疗组。实验8周后处死,测定各组小鼠血清、肝脏、脑组织中丙二醛含量及超氧化物歧化酶活性及皮肤中羟脯氨酸含量。结果与结论:与对照组相比,模型组小鼠超氧化物歧化酶活性、羟脯氨酸含量降低,丙二醛含量升高,差异有非常显著性意义(P≤0.01)。与3个治疗组相比,模型组超氧化物歧化酶活性、羟脯氨酸含量偏低,丙二醛含量偏高,且差异有显著性意义(P≤0.05),但模型组大脑中丙二醛含量与番茄红素治疗组差异无显著性意义(P>0.05)。3个治疗组相比,干细胞联合番茄红素治疗组中超氧化物歧化酶活性、羟脯氨酸含量较高、丙二醛含量较低,差异有显著性意义(P≤0.05)。提示干细胞、番茄红素具有不同程度的抗衰老作用,且二者联合的抗衰老作用优于单独使用。 BACKGROUND: Studies have demonstrated the anti-aging action of human umbilical cord mesenchymal stem cells or lycopene. However, there are few researches on the anti-aging action of their combination. OBJECTIVE: To investigate the anti-aging effects of human umbilical cord mesenchymal stem cells combined with lycopene on D-galactose-induced aging model in mice. METHODS: Subactute aging model was established in ICR mice by subcutaneous injection of D-gatactose. The experimental mice were divided into control group, model group, stem cell group, lycopene and stem cells+lycopene group. The mice were sacrificed after 8 weeks. The activity of superoxide dismutase, the content of malondialdehyde in serum, liver, brain and the content of hydroxyproline in skin were determined. RESULTS AND CONCLUSION: Compared with the control group, the activity of superoxide dismutase and thecontent of hydroxyproline were significantly reduced, but the content of malondialdehyde significantly increased in model group mice (P ≤0.01 ). Compared with three treatment groups, the activity of superoxide dismutase and the content of hydroxyproline were significantly lower, but the content of malondialdehyde significantly greater in the model group (P≤0.05). However, there was no statistical significance in content of malondialdehyde between model and lycopene groups (P 〉 0.05). The activity of superoxide dismutase and the content of hydroxyproline were significantly higher, but the content of malondialdehyde significantly lower in stem cells+lycopene group compared with stem cell or lycopene groups (P ≤ 0.05). Results suggested that human umbilical cord mesenchymal stem cells and lycopene have anti-aging effects, and the anti-aging effects of the combination of them are better than either of them.
