摘要
目的研究间充质干细胞(MSCs)对脑组织辐射损伤的修复作用,为治疗放射性脑损伤奠定理论基础。方法选取24只雄性SD大鼠随机分为3组,即对照组、模型组和治疗组。模型组和治疗组大鼠均接受直线加速器产生的X线行一次性全脑照射20Gy;正常对照组不接受照射。其中治疗组大鼠接受辐射24h、7d、14d、21d后尾静脉注射MSCs3×106/只;模型组和正常对照组大鼠尾静脉注射同体积生理盐水1mL。28d后处死所有大鼠,检测血清中丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性;采用苏木精-伊红染色法(HE染色法)制作大鼠的脑组织病理切片观察辐射对脑组织形态学的影响。结果模型组大鼠精神较差,反应迟钝,少动,毛发粗糙、变黄,眼睑分泌物增多,睁眼困难;治疗组大鼠精神好,毛发微黄,眼睑有少量分泌物。治疗组大鼠血清SOD明显高于模型组,而MDA水平低于模型组。病理结果显示模型组大脑皮质变薄,神经元细胞聚集排列,数目显著减少,可见核固缩、深染;尼氏小体融解消失,呈凋亡状态;治疗组大脑皮质较模型组明显增厚,海马神经元细胞增多,仍可见凋亡状态的尼氏小体,细胞染色较淡,损伤的细胞数目明显减少,损伤程度减轻。结论 MSCs能够使神经细胞再生,修复脑组织的辐射损伤。
Objective To study the repairing effects of mesenchymal stem cells (MSCs) on radiation induced brain injury, laying a theoretical basis for the clinical treatment of radiation injury. Methods Twenty four male SD rats were randomly divided into three groups: the normal group, model group and treatment group. Total brain irradiation with 20 Gy X -ray was performed in model group and treatment group, while no irradiation was given in the control group. Rats in the treatment group received infusion of MSCs with 3 x 106 each at 24 h, 7 d, 14 d and 21 d after irradiation, while rats re- ceived infusion of saline in model group and control groups. Rats were sacrificed on 25 day for assessing serum malondialde- hyde (MDA) concentration, superoxide dismutase (SOD) activity and pathological changes in brain tissue. Results Com- pared with control group, rats in the model group represented poor conscious, less dynamic, coarse and yellow hair, in- creased eyelid secrecions and difticult eye opening. It revealed that SOD in the treatment group was significantly higher than that in the model group, with MDA levels significantly lower ( P 〈 0. 05 ). Pathological changes in the model group were as follows : thinned cerebral cortex, neuron cells aggregation arrangement with a significant reduction in amount, nuclear pykno- sis with deep staining and melt Nissl body, showing apoptotic state. Meanwhile, significant thicker cerebral cortex and in- creased hippocampal neurons were observed in treatment group. Although apoptosis of Nissl bodies were still observed in treatment group, cells were stained lighter implying significantly reduced injury. Conclusion MSCs can effectively promote neuron regeneration and repair of radiation injury.
