摘要
目的:检测广西肝癌高发区肝细胞癌患者中血清相关差异蛋白。方法:应用基于广西肝癌高发地区为基础的病例对照研究方法,采用蛋白质芯片技术对广西肝癌高发区11例肝癌患者与11名正常健康人血清蛋白进行检测,应用ELISA法对筛检出来的蛋白质在44例肝癌血清和44名正常健康人血清中进行验证。结果:蛋白质芯片结果提示,肝癌组血清IGFBP-2的浓度为(887.48±225.91)ρg/mL,高于对照组血清IGFBP-2浓度为(558.17±125.97)ρg/mL,t=2.846 9,P=0.021 6;肝癌组血清p21的浓度为(1 040.97±395.91)ρg/mL,低于对照组(2 756.64±1 355.64)ρg/mL,t=2.570 6,P=0.013 9;ELISA结果显示,肝癌患者血清IGFBP-2浓度为(2 006.54±407.71)ng/L,高于对照组(1 463.81±634.70)ng/L,t=4.772 3,P<0.001;肝癌患者血清p21浓度为(2 293.095±1 378.03)ng/L,低于对照组(3 244.076±745.07)ng/L,t=4.026 7,P=0.000 1。结论:应用蛋白质芯片技术检测到的2个血清差异蛋白,可能是肝癌患者血清特异性生物标志,与广西肝癌高发区肝癌发生发展有关。
OBJECTIVE:To detect the differences of relative serum protein with hepatocellular carcinoma in the en- demic area of HCC in Guangxi. METHODS: To apply matching case-control study based on the endemic area of HCC in Guangxi,the serum differentially expressed proteins of the patients with hepatocellular carcinoma and the normal heahy people in the endemic area of HCC were detected by the protein chip and validated by ELISA. RESULTS: 1) Protein chips: IGFBP-2 level of HCC and level of normal heahy people were (887.48±225.91) pg/mL and (558.17±125.97) pg/mL, respectively. The levels of IGFBP-2 in HCC was higher than those in normal healty people(t=2. 846 9, P= 0. 021 6) ;p21 level of HCC and level of normal healty peopIe were (1 040.97±395.91) pg/mL and (2 756.64±1 355.64) pg/mL,re spectively. The levels of p21 in HCC was lower than those in normal heahy people(t=2. 570 6,P〈0. 013 9). 2) ELISA: Serum IGFBP-2 level of HCC was (2 006.54±407. 71) ng/L and Serum IGFBP-2 level in matched normal groups was (1 463.81±634.70) ng/L. Statistical analysis showed Serum IGFBP-2 level of HCC was higher than that of the control group(t=4. 772 3,P〈0. 001) ;Serum p21 level of HCC was (2 293. 095±1 378.03) ng/L and Serum p21 level in normal heahy people was (3 244. 076±745.07) ng/L. Statistical analysis showed Serum p21 level of HCC was lower than that of the control group (t= 4. 026 7, P= 0. 000 1). CONCLUSION:The two differentially expressed proteins detected by protein chip may be the serum liver cancer-specific biomarkers of HCC,which are related to the development of HCC in the en demic area of HCC in Guangxi.
