Optimization of fermentation conditions of fibrinolytic enzyme produced by Rhizopus chinesis sp.

摘要 研究了不同培养基对中国根霉 1 2号 (Rhizopuschinesis 1 2 )发酵生产纤溶酶的影响 ,发现C/N的降低有利于产酶 ,同时用正交实验给出了最佳培养条件为 :以麸皮水 +胰蛋白胨 +豆渣为培养基 ,接种量 2 0 % ,转速 1 70r/min ;用响应面设计确定了最佳培养基为 4.3 6661 %麸皮水 +0 .98975 %胰蛋白胨 +4 .89895 %豆渣 ;在最优条件下 ,纤溶酶的发酵效价可达 3 .0 6U/ml。发现添加金属离子Mg2 + ,Ca2 + 可以提高产量 。 The fermentation conditions of fibrinolytic enzyme produced by Rhizopus chinesis 12 were discussed . Compared with different media it was found that the decrease of C/N would facilitate the generation of fibrinolytic enzyme. The optimum fermentation conditions determined by orthogonal test and response surface experiment (Box-Behnken Designs) were as follows: inoculum size 20%, rotation speed 170 r/min and the optimum medium consisted of 4.36661 % of bran, 0.98975 % of tryptone and 4.89895 % of soybean residue. Under the optimum cultivation conditions, the fermentation titer could approach 3.06 U/ml. The output was improved by adding metal ions such as Ca 2+ , Mg 2+ . And, the changes of different parameters during fermentation were shown by curves.
