摘要
通过单因素试验和正交试验确定地衣芽孢杆菌lw-72摇瓶发酵合成纤溶酶的最佳条件,最佳培养基组成为:玉米粉0.5%、黄豆饼粉2.0%、Na2HPO4 0.4%、NaH2PO4 0.2%、CaCl2·2H2O 0.01%、KCl 0.01%;培养基初始pH为7.0;最佳培养条件:装液量30ml/250ml摇瓶,接种量10%,发酵时间为72h。在该条件下发酵液中纤溶酶酶活力为622u/ml,是初始发酵培养基的3.4倍。
The optimal shake flask fermentation conditions for fibrinolytic enzyme from Bacillus licheniformis lw-72 were determined based on single factor experiments and orthogonal experiment. The composition of optimal fermentation culture medium was as follows: corn flour 0.5 %, soybean powder 2.0 %, Na2HPO4 0.4 %, NaH2PO4 0.2 %, CaCl2-2H20 0.01%, KCl 0.01%. Fermentation was performed for 72 h in a 250 ml shake flask containing 30 ml medium at 10 % inoculum volume and initial pH value 7.0. The enzyme activity could reach 622 u/ml, a 2,4 fold higher than that under the original conditions.
