摘要
以月季良种红衣主教带腋芽茎段为外植体进行离体快速繁殖研究 .结果表明 :在 MS+2 .0 0 - 3.0 0mg· L-16 - BA+0 .10 mg· L-1NAA培养基上进行起始培养 ,其腋芽分化率达 6 3%以上 ;在 MS+2 .0 0 - 3.0 0mg· L-16 - BA+0 .10 mg· L-1NAA+1.0 0 mg· L-1GA3 培养基上进行继代增殖 ,4 5d其增殖倍数达 3.4以上 ,且长成的芽苗比较粗壮 ;增殖后的芽苗在 1/ 2 MS(大量元素和琼脂用量减半 ,呈半液体状态 ) +0 .50 - 1.0 0mg· L-1NAA或 0 .50 - 0 .75mg·L-1IBA培养基上培养 ,根系生长良好 ,30 d小苗发根率达 90 %以上 ;经炼苗后移栽在以沙壤土、砻糖灰和蛭石混合 (体积 1∶ 1∶ 1)基质中 ,成活率达 80 %以上 .当小苗长到 10 cm左右移植于田间栽培 ,90 d后可陆续现蕾 ,180
Stems with axillary buds from Cardinal, elite of China rose, were used as explants for in vitro rapid propagation. The results showed that in primary culture on the medium MS+2.00-3.00 mg·L -1 6 BA+0.10 mg·L -1 NAA, percentage of axillary bud differentiation was higher than 63%. Then in subculture on the medium MS+ 2.00 -3.00 mg·L -1 6 BA+0.10 mg·L -1 NAA+1.00 mg·L -1 GA 3, the multiple of regeneration was more than 3.4 in 45 days. And the grown shoots were sounder. Well grown shoots rooted on the medium 1/2 MS (half of the amount of macroelements and agar)+0.50-1.00 mg·L -1 NAA (or 0.50-0.75 mg·L -1 IBA), and rootability was higher than 90% in 30 days. Eventually, the plantlets were transplanted to the substrate made of sandy loam, rice hullash and vermiculite (1∶1∶1) and the survival rate was higher than 80%. When their height grew up to 10 cm, they were again transplanted to the field, alabastrums appeared 90 days later and normal blooming began after 180 days.
