原发性肝细胞癌细胞中金属硫蛋白的表达及其与细胞凋亡相关性的研究 被引量:4

摘要 目的:确定原发性肝细胞癌(Primary Hepatocellular Carcinoma HCC)组织中金属硫蛋白(Metallothionein MT)的表达水平及其与细胞凋亡的关系,探讨MT在人类肿瘤细胞中表达的生物学意义及临床应用价值。方法:采用多克隆抗体免疫组化染色及细胞凋亡的末端标记技术,检测了20例HCC组织和16例正常人肝组织及肝癌周围正常及肝硬变组织中的MT表达及凋亡细胞(Apoptotic cell APC)数。结果:正常及肝癌周围肝组织中有高的MT表达及低APC数;20例HCC组织中有12例(60%)未显示MT染色,其余8例也只显示低度的MT染色,而HCC组织的APC数明显高于正常及肝癌周围肝组织。结论:HCC组织中的MT含量与正常及肝癌周围肝组织相比明显降低而APC数则增高,MT表达与APC数呈负相关。 Objective To confirm the pattern of Metallothionein MT expression in hepatocellular carcinoma HCC and the correlation of the number of apoptotic cell APC with MT expression. To explore the biological significance of MT expression in human tumors and to evaluate its clinical application. Methods The MT expression was determined by immunohistochemical staining with polyclonal antibody and a number of APC by Tunel technique. Results Normal liver had consistent MT staining with very low incidence of APC. Twelve of 20 HCC cases 60% showed no MT staining and the rest also showed a low grade MT staining as compared with normal liver or HCC-surrounding liver which included normal or cirrhotic liver. The incidence of APC in HCC was markedly higher than that in normal liver or HCC-surrounding liver tissues. Conclusion It has a negative correlation between the number of APC and MT expression.
