摘要
目的 对脑血管内皮细胞中神经肽 Y(NPY)的 Y1 受体进行精确定位 .方法 用针对 NPY的 Y1 受体 C末端肽片段的一种纯化的抗血清 ,用免疫组织化学方法和包埋前免疫金 -银加强法观察 Y1 受体在脑血管内皮细胞中的分布 .结果 大脑皮质和髓质、丘脑、下丘脑、小脑和脑干的微血管有散在的含 Y1 受体免疫反应的 (- Ir)内皮细胞 ,孤立的脑底动脉及其分支的一些内皮细胞也具有 Y1 受体免疫反应性 (- L I) ,Y1 受体 - L I分布于内皮细胞的血管腔面和平滑肌细胞面的细胞膜和胞质中 .结论 来源于血液、内皮细胞和血管周围神经的 NPY通过内皮细胞上 Y1 受体的介导 。
AIM To accurately localize Y_1 receptor of neuropeptide Y(NPY) in the endothelium of cerebral blood vessels. METHODS An affinity-purified antiserum against a C-terminal peptide fragment of Y_1-R was used with the immunobiochemistry and pre-embedding electron gold-silver immunostaining. RESULTS Disperse Y_1-R-Ir endothelium wasobserved in the micro-blood vessels of the brain cortex and brain medulla, campus, hippocampus, cerebellum and brainstem, and also in the isolated basal cerebral arteries and their branchs. There was Y_1-R-LI in the plasmalemma and plasma facing the smooth muscle cells and the lumen of the blood vessels. CONCLUSION NPY from the blood, endothelium and the nerve around the blood vessels may play a role in the regulation of cerebral blood flow and stimulate the proliferation of the smooth muscle cells via Y_1-R.
