摘要
目的了解河南省与手足口病相关的埃可病毒1型(echovirus 1,E1)河南分离株2140/Henan/2010的基因组特征,对其进行全基因测序,并与其他肠道病毒基因进行同源性比较和进化分析。方法采用RT-PCR扩增全基因,采用SeqMan将所测的全部序列进行拼接,使用MegAlign进行同源性比较,MEGA4进行进化分析。结果 2140/Henan/2010基因组全长7 325bp。与原型株Farouk比较,编码区氨基酸同源性为97.6%,核苷酸同源性为80.7%,其中P1核苷酸差异为21.2%,VP1核苷酸差异为16.5%,P2、P3核苷酸差异分别为19.9%、22.4%。进化分析发现,2140/Henan/2010和E1/Farouk仅在P1区和VP1区形成同一分枝,2140/Henan/2010和E1/Farouk在P1和VP1区、P2区、P3区分别与E4、E30、E12亲缘关系最相近。结论与原型株相比,2140/Henan/2010株编码区氨基酸序列差异很小,核苷酸变异较大,核苷酸变异多为无义突变,不影响氨基酸序列的变化;推测E1在P1以外的多个基因组区段分别与其他血清型肠道病毒发生重组。
Objective To sequence the whole genome of Echovirus 1 (2140/Henan/2010 strain) and compare with other enteroviruses. Methods The whole genome was sequenced by RT-PCR; sequence assembly, nu- cleotide and amino acid sequence analysis, and phylogenetic analysis were done by using software SeqMan, MegAlign and MEGA4. Results The genome of 2140/Henan/2010 strain was 7 325 nucleotide long that shared 80.7% and 97.6%homology in nucleotides and deducted amino acids, respectively with the prototype strain E1/Farouk. 2140/Henan/2010 and E1/Farouk are clustered into the same evolutionary group and both strains are most closely related to E1 are E4, E30, and El2 in the Pl, P2, and P3 regions, respective- ly. Conclusions Comparing with the prototype strain, the coding region of 2140/Henan/2010 strain has a si- lent mutation but most nucleotide variations do not bring changes in the corresponding amino acids. Different gene fragments other than P1 region of E1 may recombine with different enterovirus serotypes.
