摘要
目的:分析与黄连清热燥湿、泻火解毒对应的5种药效组分变化规律,探讨饮片配伍、复方、剂型与药效组分的相关性,为新药的研发和质量评价体系的建立提供科学数据。方法:采用单味药、药对和复方及其制剂对应的方法制备样品并进行对照,采用RP-HPLC法测定黄连5种药效组分的含量。结果:与黄连功能对应的药效组分是盐酸药根碱、盐酸表小檗碱、盐酸黄连碱、盐酸巴马丁、盐酸小檗碱,其含量依次为2.18±0.37、8.81±0.55、18.57±0.35、7.50±0.12、38.41±1.26 mg·g^(-1)。结论:黄连药效组分盐酸药根碱-盐酸表小檗碱-盐酸黄连碱-盐酸巴马丁-盐酸小檗碱,其清热解毒组分是1:3.2:4.88:2.72:12.85;其清热燥湿组分是1:2.84:4.62:2.32:11.3;其清热泻火组分是1:4.21:6.76:3.39:16.87;且各药效组分之间有显著性差异(P<0.01)。黄连的功能与配伍和剂型相关,黄连的功能物质是药效组分。
Objective ： To analyze the variation of the five active component alignments （ ACA ） corresponding to the basis functions of Rhizoma Coptidis which were clearing away heat and eliminating dampness and purging fire and toxic material.To study the correlation of compatibility of medicines, formulation and active components alignments.To provide scientific data for the discovery of new traditional Chinese medicine and the establishment of its quality evaluation methods. Methods ： The samples of Rhizoma Coptidis and its prescriptions were prepared based on the classic Chinese medicine preparation method.The five ACA of Rhizoma Coptidis were determined by reversed phase high-performance liquid chromatography （ RP-HPLC ） compatibility. Results ： The ACA corresponding to the basis functions of Rhizoma Coptidis were Hydrochlaride gatrorrhizine, Hydroehloride epiberberine, Hydrochloride coptisine, Hydroehloride palmatine, Hydroehloride berberine.The contents of those ACA were respectively （ 2.18 ±0.37 ）mg · g^-1（ 8.81 ±0.55 ） mg · g^-1, （ 18.57 ± 0.35 ）mg · g^-1, （ 7.50 ± 0.12 ） mg · g^-1, （ 38.41 ±1.26 ） mg · g^-1 . Conclusions ： The ACA of Rhizoma Coptidis were Hydrochloride gatrolThizine, Hydrochloride epiberberine, Hydrochloride coptisine, Hydrochloride palmatine, Hydrochloride herberine.The ACA proportion of clearing away heat and toxic material were 1 ： 3.2 ： 4.88 ： 2.72 ： 12.85.The ACA proportion of heat-dampness were 1 ： 2,84 ： 4.62 ： 2.32 ： 113. The ACA proportion of eliminating heat and purging fire were 1 ： 4.21 ： 6.76 ： 3.39 ： 16.87.There were high significant differences among all functions （ P〈0.01 ） .The active components alignments were correlated with compatibility of medicines and formulation.The functionality substance of Rhizoma Coptidis were the active component alignment.
