JNK及P38信号分子在周期性张应变下调大鼠椎间盘纤维环细胞Aggrecan mRNA表达中的作用 被引量:1

摘要 [目的]探讨JNK及P38信号分子在周期性张应变下调椎间盘纤维环细胞蛋白多糖-aggrecan mRNA表达中的作用。[方法]采用自制周期性张应变加载装置,对培养的大鼠椎间盘纤维环细胞施以幅度为10%,频率为1Hz的张应变刺激,Western blot分析周期性张应变处理不同时间对椎间盘纤维环细胞MAPK家族中JNK及P38信号分子磷酸化水平的影响,并探讨其特异性阻断剂对周期性张应变下调椎间盘纤维环细胞Aggrecan mRNA表达的影响。[结果]周期性张应变可上调纤维环细胞JNK蛋白磷酸化水平,具有双向、快速的特点,但对P38磷酸化水平无明显影响;JNK抑制剂SP600125预处理可部分逆转周期性张应变下调纤维环细胞aggrecan mRNA的作用。[结论]周期性张应变可激活椎间盘纤维环细胞JNK信号分子,并通过该信号分子下调椎间盘纤维环细胞aggrecan mRNA的表达。 [Objective] To investigate the role of JNK and P38 signaling in the down -regulation of aggrecan mRNA in- duced by cyclic tensile strain in annulus fibrosus cells. [ Methods] Annulus fibrosus cells cultured in a monolayer were treated with 10% CTS at 1Hz by using a custom - built device. Western blot analysis was performed to detect JNK and P38phosphorylation levels. A specific inhibitor was added to the cell culture. Subsequently, aggrecan mRNA expression induced by CTS was assayed byRT-PCR. [Results] JNK, but not P38, was activated by CTS with a rapid and biphasic time course; treatment with SP600125 (a JNK inhibitor) prevented CTS- induced activation of JNK in the annulus fibrosus cells. Moreover, SP600125 partially reversed the effects of CTS on aggrecan mRNA expression. [ Conclusion] JNK, but not P38, signaling me- diates, at least in part, the down - regulation of aggrecan mRNA induced by CTS in annulus fibrosus cells.
