摘要
为了建立中药闹羊花中Rhodojaponin-III(Rj-III)和Rhodojaponin-VI(Rj-VI)的含量测定方法,本文采用高效液相色谱-蒸发光散射检测方法(HPLC-ELSD)直接测定闹羊花75%丙酮水提取物中Rj-III和Rj-VI的含量。采用Agilent ZORBAX SB-C_18(4.6 mm×250 mm,5μm)色谱柱,以甲醇-水(40∶60)为流动相,流速1 mL/min,柱温30℃,ELSD(Varian 380-L)检测器,雾化器温度55℃,漂移管温度80℃,气体压力为25 psi,载气速度为1.6 L/s,增益为500。结果表明,闹羊花中Rj-III和Rj-VI的含量分别为1.21 mg/g和1.03 mg/g;Rj-III和RjVI回归方程分别为Y=2026.8X-789.2,R^2=0.9979;Y=3777.6X-1849.5,R^2=0.9980;分别在0.4~2.83μg/μL和0.6~4.2μg/μL范围内,进样量与峰面积呈良好的线性关系,该方法简单可行,快速,精密度、重现性和稳定性均较好,可应用于中药闹羊花中Rj-III和Rj-VI的含量测定。
The objective of this study is to establish a HPLC-ELSD method for the quantitative determination of Rhodoja- ponln-Ⅲ (Rj-Ⅲ) and Rhodojaponin-Ⅵ (Rj-Ⅵ) in traditional Chinese medicine Rhododendron molle. The content of Rj-Ⅲ and Rj-Ⅵ in 75% acetone-water extract of R. molle was determined by HPLC-ELSD using the foflowing condi- tions,column:Agilent ZORBAX SB-C18(4.6 mm ×250mm,5μm) ,mobile phase:methanol-water (40: 60) ,flow rate: 1 mL/min, column temperature :30 ℃, detector: ELSD ( Varian 380-LC ), atomization temperature :55 ℃, drift tube tem- perature:80 ℃ ,gas pressure:25 psi,flow rate of carrier gas:1. 6 L/s,gain:500. The result showed that Rj-Ⅲ and Rj-Ⅵ had a good linearity between 0.4-2.83 μg/μL ( Y = 2026. 8X-789. 2,R2 = 0.9979) and 0.6 -4.2 μg/μL ( Y = 3777.6X-1849.5 ,R2 = 0. 9980 ) , respectively. Finally,the content of Rj-Ⅲ and Rj-Ⅵ were determined to be 1.21 and 1.03 mg/g, respectively. Therefore ,the method developed in this study was simple, fast, accurate, reproducible and accu- rate. It can be used for the quantification of Rj Ⅲ and Rj Ⅵ in R. molle.
